Objective Worldwide,lung cancer cases and deaths are rising.Most patients with lung cancer are at an advanced stage at diagnosis and therefore have a poor prognosis,with an overall 5-year survival rate of 10% to 15%.As a heterogeneous disease,lung cancer includes a variety of pathological and clinically relevant subtypes,of which non-small cell lung cancer is the most common subtype,including adenocarcinoma,squamous cell carcinoma,and large cell lung cancer,while adenocarcinoma is now the main histological type in lung cancer patients,accounting for almost half of all lung cancer cases.Despite the achievements in the pathogenesis of lung adenocarcinoma and the development of new treatments,lung adenocarcinoma unfortunately remains one of the most aggressive and rapidly fatal tumor types,with an overall survival of less than 5years.Therefore,it is particularly important to further explore the new pathogenesis and treatment of lung adenocarcinoma.The aim of this study is to investigate the relationship between the expression of ATAD2 and the prognosis and immune infiltration of lung adenocarcinoma(LUAD),and to provide a new theoretical basis for exploring new prognostic biomarkers and possible therapeutic targets of LUAD.Methods A case-control study was conducted.Serum samples from 40 patients with lung adenocarcinoma and 34 healthy subjects were collected from the Affiliated Hospital of Dali University.ELISA was used to compare the expression of ATAD2 in serum samples of lung adenocarcinoma and normal people,and the expression of ATAD2 was verified by bioinformatics analysis.The relationship between prognosis and immune infiltration of lung adenocarcinoma was also investigated.Results(1)The serum level of ATAD2 in lung adenocarcinoma group was higher than that in control group,and ATAD2 was highly expressed in lung adenocarcinoma.(2)ATAD2 is highly expressed in a variety of tumor tissues,such as bladder transitional carcinoma,breast invasive carcinoma,cholangiocarcinoma,colon cancer,esophageal cancer,hepatocellular carcinoma,lung adenocarcinoma and lung squamous cell carcinoma.(3)High ATAD2 expression was associated with poor overall survival(OS:HR=1.5,P=0.0056)in lung adenocarcinoma.(4)The high expression of ATAD2 was related to gender,age,race,stage and TP53 mutation status.(5)ATAD2 expression was positively correlated with tumor purity and infiltration of CD8 + T cells,neutrophils,dendritic cells and macrophages.It was negatively correlated with the infiltration level of B cells and CD4 + T cells.(6)ATAD2 was significantly enriched in PLK1 pathway,FOXM1 pathway,ATR pathway,AURORA B pathway,and histone deacetylase pathway.Conclusion ATAD2 is highly expressed in lung adenocarcinoma,and high ex pression of ATAD2 is associated with poor overall survival of lung adenocarcino ma.ATAD2 can be used as a potential biomarker for early diagnosis and prognos is of lung adenocarcinoma.ATAD2 is related to immune infiltration and can be u sed to evaluate the level of immune cell infiltration in tumor tissues.ATAD2 ma y play a role in the occurrence and development of tumors by regulating the pr oliferation of tumor cells,regulating DNA damage repair,participating in DNA r eplication and cell cycle progression.
