Cerium is the most abundant rare earth element in the crust,mainly in the form of Cerium(IV)oxide(Ce O2).Ce O2 has attracted widespread attention due to its unique 4f electrons.The fluorite phase structure of Ce O2’s face centered cubic lattice can affect the number of oxygen vacancies due to changes in oxygen concentration and environmental temperature.This special structure has great potential and broad application prospects in catalysis,optics,electricity,and magnetism.Nanozymes are nanomaterials with enzymatic properties.As an artificial synthetic enzyme,nanozymes have the advantages of low synthesis cost,high catalytic activity,simple preparation process,and easy storage,overcoming many limitations of natural enzymes.Due to their extensive potential applications,they have become an emerging field connecting nanotechnology and biology,such as Alzheimer’s disease(AD),tumor research,Parkinson’s disease,enteritis,and so on.AD is a complex neuro Degenerative disease.With the aging of the population,the prevalence of AD is expected to continue to increase.AD and its related research are still hot and difficult topics in current research.This article synthesized a new type of Ce O2-x using hydrothermal method.The TEM results show that the particle size is approximately 5 nm.Through X-ray photoelectron spectroscopy,X-ray powder diffraction,element surface analysis,oxygen vacancy test,zeta potential and other data analysis,Ce O2-x is a cubic fluorite structure,with a Ce3+content of 14.44%.It has a high lattice defect and a high activity of Catalase like and Superoxide dismutase like enzymes.Further treatment of E.coli OP50 with Ce O2-x nanozyme revealed that Ce O2-xnanozyme had no effect on the growth of E.coli OP50.Therefore,subsequent experimental results were not related to the growth of E.coli OP50.Subsequent experiments have shown that 0.05 mg/m L Ce O2-x nanozyme can prolong the lifespan of CL4176 and alleviate their paralysis.After treatment with 0.05 mg/m L Ce O2-xnanozyme,the average lifespan of CL4176 was extended by 5.08%and PT50 was extended by 1.23 hours.Ce O2-x nanozyme can significantly reduce the reactive oxygen species in the body of CL4176,resulting in a 13.2%decrease in reactive oxygen species levels.At the same time,Ce O2-x Nanozymes can significantly reduce the intracellular Amyloidβlevels.The RNA seq sequencing results showed that Ce O2-x Nanozymes can upregulate genes related to collagen production such as col-37,col-40,and col-2.Collagen has a protective effect on neurons while reducing oxidative stress response;Ce O2-x Nanozymes can downregulate the sym-2 gene,reducing sym-2 expression can alleviate neuromuscular atrophy caused by neurodegenerative diseases.In conclusion,a new Ce O2-x nanozyme with high Ce3+content,oxygen vacancies and double enzyme activities like Catalase and Superoxide dismutase was synthesized by hydrothermal method.And it was found that it can reduce the levels of reactive oxygen species in organisms,reducing the production of Amyloidβalleviates the paralysis rate of CL4176 and prolongs its average lifespan.This study provides new ideas and experimental basis for the treatment of Alzheimer’s disease in the future,and provides a certain reference for new drug research.
