| Objective: In this study,we analyzed the relationship between SGK1 gene polymorphisms and primary hypertension in Mongolian population to explore whether there is a subtype of hypertension caused by SGK1 gene mutations in hypertension,and to provide a basis for accurate diagnosis and treatment of hypertension.METHODS:(1)Systematic review and bibliometrics: The published research literature on SGK1 and hypertension between January1992 and April 2022 was searched in Web of Science,parameters such as complete records and cited references were extracted,and bibliometric methods were used to analyse the publication profile and identify subject hotspots and frontiers.(2)Case-control study:From the Mongolian population in the cross-sectional study of the previous NSF project,372 cases and199 controls were finally included according to the inclusion and exclusion criteria of the case and control groups.Data were entered using Epidata 3.0 software,single nucleotide polymorphisms of SGK1 gene were detected using multiplex ligase conformation reaction technique,the relationship between Hardy-Weinberg equilibrium and alleles and hypertension was analysed using Haploview software,and the relationship between genotype and hypertension was analysed using SNPstats online analysis system and SPSS25.0 statistical analysis software.Results:(1)(1)There were 282 SGK1-related hypertension publications at the time of the search,with China,Germany,Japan and the United States leading the way in this area of research.(2)The University of Tübingen,the University of California,San Diego and the University of Lausanne are the most influential institutions in this field.(3)The hotspot of research on SGK1-related hypertension focuses on the mechanism of hypertension development and target organ damage,and the frontier of research is the mechanism of hypertension development and SGK1 inhibition.(2)(1)The rs1057293,rs147657480,rs148941985,rs1743966 and rs55957552 loci of the SGK1 gene are all consistent with Hardy-Weinberg equilibrium.(2)The rs55957552 locus of the SGK1 gene was associated with primary hypertension in Mongolians.(3)Stratified analysis showed that the variation in rs55957552 was statistically associated with the prevalence of hypertension in the middle to high school population with education level,P=0.03.(4)In haplotype association analysis,two SNP loci,rs1057293 and rs1743966,forming three haplotypes,GA,AG and GG,were not associated with the prevalence of essential hypertension in Mongolians.(5)SGK1 expression was not detected at the protein level.CONCLUSION: In a systematic review of bibliometrics,an association was found between the SGK1 gene and the development of hypertension.At the genetic level,the SGK1 gene rs55957552 is a risk factor for Mongolian primary hypertension.At the protein level,SGK1 expression was not detected in urinary exosomes. |
