| Objective:This study aims to investigate the effect of secukinumab treatment on psoriasis proteomics,screen out the key differential proteins and key signaling pathways,further reveal the related mechanisms of secukinumab treatment in psoriasis,and provide new targets for the treatment of this disease.Methods:According to the inclusion and exclusion criteria,for five patients who met the criteria,venous blood was taken before and after 5 weeks of medication,which was secukinumab injection at 0,1,2,3 and 4 weeks,300 mg per week,2 doses of subcutaneous injection.Separation of the serum from the collected whole blood,serum was collected for the Label-free proteomics experiments,after library search and protein quantification,then we selected the key differentially expressed proteins associated with psoriasis,then by GO(Gene ontology)and KEGG(Kyoto Encyclopedia of Genes and Genomes)for the related functional enrichment analysis of the selected differentially expressed proteins.Results:Serum proteomic analysis before and after secukinumab treatment for psoriasis,in the present study,a total of 17 differentially expressed proteins were selected,of the 13 upregulated differentially expressed proteins,respectively,are the Q12907(LMAN2),O43157(PLXNB1),O14960(LECT2),Q13508(ART3),Q66K66(TMEM198),P01619(IGKV3-20),P0DOX5,P13796(LCP1),P80748(IGLV3-21),Q8NF91(SYNE1),P10451(SPP1),Q8IUC6(TICAM1),P01703(IGLV1-40).Four downregulated differentially expressed proteins,they are A0A0A0MT89(IGKJ1),Q9NZP8(C1RL),P01130(LDLR),and P51654(GPC3).GO functional annotation and enrichment analysis showed that differentially expressed proteins were focused in immune system processes,in cellular component analysis,in extracellular regions and in molecular functions,differentially expressed proteins were focused on protein-containing complex binding.KEGG pathway annotation and enrichment analysis showed that four disease-related pathways were selected,including Toll-like receptor pathway,Hepatitis C pathway,Lipid and atherosclerosis pathway,and Human papillomavirus infection pathway.Conclusion:Through the serum proteomic analysis of psoriasis patients,17 differential expressed proteins related to the pathogenesis of psoriasis and 4 related ways of psoriasis,further revealing the related mechanism of secukinumab injection treatment of psoriasis,and providing the molecular biological basis for the precise targeted therapy of the disease. |
