| Objctive:Detection of autotaxin(ATX)and transforming growth factor-β2(TGF-β2)levels and cytomegalovirus in aqueous humor In the presence(positive,positive,+)or absence(negative,-)of Cytomegalovirus(CMV),they were discussed separately with intraocular pressure(IOP).Correlation between levels and the number of corneal endothelial cells(CEC).Method:This topic is divided into two parts.The first part is to analyze the levels of ATX and TGF-β2 in aqueous humor in patients with PS and their relationship with IOP.Aqueous humor samples from PSS patients who were confirmed in the People’s Hospital of Inner Mongolia Autonomous Region and received PCR test for aqueous humor samples were collected;Polymerase chain reaction(PCR)was used to detect CMV in aqueous humor Deoxyribonucleic acid(DNA),4 3 cases of CMV-positive P were selected SS patients were in the CMV+group,and another 35 cases were CMV negative PSS Patients as the CMV-group.An additional40 patients who required cataract surgery without other eye diseases and systemic diseases were selected as the control group.The levels of ATX and TGF-β2 in aqueous humor were detected and compared by the double antibody sandwich E LISA method,and intraocular pressure was measured by noncontact tonometer.To explore the correlation between the episodes of PSS,their respective relationship with CMV DNA,and whether it affects intraocular pressure.The second part analyzes the level of TGF-β2 in aqueous humor in relation to the number of corneal endothelial cells.The number of corneal endothelium in the affected eye is measured with a non-contact corneal endothelial microscope.Correlation analysis of TGFβ2 levels in aqueous humor with the number of corneal endothelial cells.Outcome:1.Compared with IOPs and CEC count between CMV+group and CMV-group,there was no significant difference in IOP(t=0.361,P>0.05),and the number of corneal endothelial cells in the CMV-group was significantly higher than that in the CMV+group,and the difference was statistically significant(t=-2.245,P<0.05).2.The ATX levels in the CMV+group and the CMV-group were significantly higher than those in the control group,and the difference was statistically significant(P<0.001),and the ATX level in the CMV+group was significantly higher than that in the CMV+group(P<0.001).The TGFβ2 level in the CMV+group was significantly lower than that in the CMV-group(P<0.05),and the TGFβ2 level in the CMV+group and the CMV-group was significantly higher than that in the control group,and the difference was statistically significant(P<0.05,P<0.001).3.ATX and TGF-β2 were not correlated in either group(CMV+group:r=-0.031,P>0.05;CMVgroup:r=0.116,P>0.05;control group:r=0.126,P>0.05).4.In three groups,ATX levels were positively correlated with intraocular pressure(CMV+group:r=0.447,P<0.01;CMV-group:r=0.302,P<0.05;control group:r=0.365,P<0.05;),while TGFβ2 and intraocular pressure were not correlated(CMV+group:r=0.047,P>0.05;CMV-group:r=0.196,P>0.05;control group:r=0.243,P>0.05).5.In three groups,There was a positive correlation between TGFβ2 level and the number of corneal endothelial cells(CMV+group:r=0.314,P<0.05;CMV-group:r=0.324,P<0.05;control group:r=0.366,P<0.05)6.Univariate linear regression analysis of intraocular pressure in patients with PSS showed that the effect of ATX level on intraocular pressure was statistically different(b=0.019,t=3.215,P<0.001,and there was no significant difference in the effect of CMV infection on intraocular pressure(b=-0.716,t=-0.361,P>0.05).7.Multivariate linear regression analysis of the number of corneal endothelial cells in PSS patients showed that TGF β2 levels had a statistically different effect on the number of corneal endothelial cells independently of age,CMV infection and other factors(b=0.118,t=2.766,P<0.01).Conclusion:1.ATX and TGFβ2 concentrations in aqueous humor were associated with PSS,and ATX increased with CMV infection.TGFβ2 decreases with CMV infection.2.ATX concentration in aqueous humor was positively correlated with intraocular pressure in PSS patients;The concentration of TGFβ2 in aqueous humor was positively correlated with the number of corneal endothelial cells in PSS patients.3.There is no correlation between ATX and TGFβ2 concentrations in aqueous humor,although the role and site of the action are similar.4.The effect of ATX on intraocular pressure is not interfered with by CMV and can be used as a new target for intraocular pressure control.5.TGFβ2 is independent of CMV infection,age,etc.,has an effect on corneal endothelial cells,and is likely to be a protective factor,but in vitro studies are needed to verify its mechanism. |
