| Objective to investigate the expression of lncRNA RP11-400N13.2 in gastric cancer cells and serum and its diagnostic value for gastric cancer,and to study the effect of lncRNA RP11-400N13.2 on gastric cancer cells and its regulatory mechanism.Methods the expression of RP11-400N13.2 in three gastric cancer cell lines(MGC-803,BGC-823,SGC-7901)and normal gastric epithelial cells(GES-1)was detected by RT-PCR,to verify the expression of RP11-400N13.2 and its association with Clinicopathological features,to draw ROC curves to analyze its diagnostic value,and to analyze the targeted binding relationship between RP11-400N13.2 and miR-940 by bioinformatics websites,the expression of miR-940 was verified in gastric cancer cell lines.Results (1)the expression of RP11-400N13.2 in gastric cancer cell line and serum was low(P <0.001),and it was related to distant metastasis(P= 0.0044),the ROC curve showed that the area under the curve of RP11-400N13.2 was 0.765(P<0.001),which suggested that RP11-400N13.2 had diagnostic value for gastric cancer.(2)the results of database analysis showed that miR-940 was highly expressed in gastric cancer and might have a targeted binding relationship with RP11-400N13.2.(3)the expression of miR-940 in gastric cancer cell line was significantly increased by RT-PCR(P<0.001).(4)Overexpression of RP11-400N13.2 significantly reduced miR-940 expression in gastric cancer cells.Conclusions the lncRNA RP11-400N13.2 is low expressed in gastric cancer cells and serum of patients,and it is related to distant metastasis.Mir-940 is highly expressed in gastric cancer. |
