| Ganoderma calidophilum is one of the unique Ganoderma species in Hainan Island,commonly known as "Ganoderma king".It is used for health care and treatment of stomachache by Li compatriots.Because of its small size and low yield,it can not be cultivated in large quantities,and the price is very expensive.Up to now,there are few studies on the chemical constituents of Ganoderma calidophilum.In order to further explore the active constituents of Ganoderma calidophilum,understand the chemical structure diversity of Ganoderma calidophilum,and clarify the pharmacodynamic material basis of Ganoderma calidophilum,this paper took Hainan wild Ganoderma thermophilum as the object,identified its ethanol extracts,and evaluated their biological activities on this basis,so as to provide reference for the future research It lays a foundation for the innovative utilization of Ganoderma calidophilum wild resources.The main results were as follows:Chemical constituents were isolated from the fruiting bodies of Ganoderma calidophilum by Thin layer chromatography,normal phase silica gel column chromatography,reversed phase C18 column chromatography and other column chromatography techniques and HPLC method.Thirty-one compounds were isolated from Ganoderma calidophilum and were identified,including 26 triterpenoids,3 heteroterpenoids and 2 phenolic compounds.There were 18 new compounds as: ganodecalone C-L,ganocalidin G,(24E)-3,11-dioxolanosta-8,24-dien-26-oic acid,3,11-dioxo-lanosta-8,25-dien-24(R),26-diol,(24E)-lanosta-8,24-dien-3,11-dione-26-al,3,11-dioxo-lanosta-21,24-epoxy-8-en-26-al,3,11-dioxo-lanosta-21,24-epoxy-8-en-26-dimethoxy,3,7,11,15,23-pentaoxolanosta-8,20E(22)-dien-26-oic acid methyl ester。The inhibitory activities of α-glucosidase,acetylcholinesterase and antitumor activities of 31 compounds were determined.The results showed that compounds 11-oxo-lucidadiol,ganodermic acid T-Q,ganodecalone F,ganoderic aldehyde A,(24E)-lanosta-8,24-dien-3,11-dione-26-al,ganoleucin C and 11β-hydroxy-lucidadiol had inhibitory activity on α-glucosidase,especially ganodermic acid T-Q and(24E)-lanosta-8,24-dien-3,11-dione-26-al,and the IC 50 values are 1.552 and 1.032 μM;compounds ganoleucin C and ganotheaecolumol H had inhibitory activity on acetylcholinesterase,and the IC 50 values are4.58 and 5.36 μM;all the 12 compounds have antitumor activities.The inhibition of ganodermic acid T-Q on BEL-7402,ganoderic aldhyde A,(24E)-lanosta-8,24-dien-3,11-dione-26-al on K562 and ganodecalone I on Hela were the most prominent,with IC 50 values of 11.35,5.59,5.79 and 12.82 μM respectively. |
