| Folium isatidis-one of the herbs commonly used in Chinese medicine,is the dried leaf of Isatis indigotica Fort.It was first recorded in the 《Mingyi Bielu》,and was included in the first part of the《Chinese Pharmacopoeia》(2000).It has the effect of releasing heat and detoxifying.It is known as the top grade of clearing heat and detoxifying.It specializes in warming pathogenic heat diseases,excess heat accumulation,and carbuncle,ulcer,swelling and toxin.It can be taken and applied externally.It is widely used.Its chemical components mainly include alkaloids,organic acids,etc.Its pharmacological effects include anti-virus,antibacterial,anti endotoxin,anti-tumor,etc.In this thesis,the chemical components of Folium Isatidis were studied.18 Kg dried medicinal materials of Folium Isatidis were crushed,heated and refluxed with 75% ethanol for extraction.The obtained precipitation was extracted with petroleum ether,ethyl acetate and n-butanol in turn.And the antipyretic effect of each extraction site on the fever model of rats induced by lipopolysaccharide was also studied,and the antipyretic effect was n-butanol extraction site > ethyl acetate extraction site > petroleum ether extraction site.The n-butanol extraction site was separated and purified with macroporous resin,silica gel,Sephadex LH-20,ODS and preparative liquid chromatography.Then,Molecular structure(compound 2)the compounds were identified according to the physical and chemical properties and spectral data.thirty-seven compounds were isolated and identified from the n-butanol fraction of Folium Isatidis as(Z)-9-heptadecenoic acid(1),5-hydroxy-2-indolinone(2),3-hydroxy-3-(2-hydroxyethyl)indolin-2-one(3),nicotinamide(4),2′-O-Methyluridine(5),safowerine A(6),thymidine(7),(7R,8S)-erythro-guaiacylglycerol(8),4-hydroxyindole-3-acetonitrile(9),2,6-dimethoxy-4-hydroxy-phenol-1-O-β-D-glucopy ranoside(10),Isotachioside(11),tachioside(12),Hypoxanthine(13),Indoline-2-one-5-O-β-D-glucopyranoside(14),adenosine(15),6-hydroxylindole-3-carboxylic acid-6-O-β-D-glucopyranoside(16),3-Acetamido-indole sulfonamide(17),(3S,5R,6R,7E,9S)-megastigman-7-ene-3,5,6,9-tetrol 3-O-β-D-glucopyranoside(18),transtorine(19),phenylalanine(20),L-tryptophan(21),anoectochine(22),6-O-β-Dglucopyranosy-β-D-(1-O-sinapoyl,2-O-feruloyl)glucopyranose(23),2-(acetylamino)benzoic acid(24),sinapic acid(25),4-O-sinapoyl-a/β-D-glucopyranose(26),6,6’-diO-sinapoylsurcose(27),3S,2′S,7′′S,8′′R-Isatindigotindoline E(28),3R,2′R,7′′R,8′′SIsatindigotindoline E(29),6-O-β-D-glucopyranosy-β-D-(1-O-sinapoyl-2-O-sinapoyl)glucopyranose(30),Dibutylterephthalate(31),indole-3-acetonitrile(32),isatindigotindoline A(33),wrightiadione(34),4-Phenyl-3-Butenoic Acid(35),NButyl-N’-(2’-nitrophenyl)urea(36),Tryptophan(37).One was isolated for the first time from the Isatis indigotica Fort,twenty-nine compounds were all isolated from Isatis indigotica Fort and this plant for the first time.Twenty-two alkaloids,including one new natural product(compound 36),four new compounds(compounds 9,17,28,29),thirteen organic acids(Compounds 1,8,10-12,23-27,30-31,35),one monoterpene(Compound 18),and one other type(Compound 34).The four newly discovered compounds all belong to indole alkaloids,and indole alkaloids have good biological activities against tumors and viruses,which are important references for the later clinical development of new drugs.This experiment enriched the chemical composition of Isatis indigotica Fort and provided quality markers for Isatis indigotica Fort and its adult formulations,and provided a reference base for the study of pharmacological activity of Isatis indigotica Fort. |
