Background and objective:Ovarian cancer(OC)is one of the most common gynecological malignancies with highest mortality in three malignancies.OC can be divided into epithelial tumors,sex cord-stromal cell tumors,germ cell tumors and other types of tumors according to its pathological types.Epithelial ovarian cancer(EOC)was the most common type of ovarian cancer,accounting for about 90%.Due to the fact that the ovary is located deep in the pelvic cavity,with no peritoneal coverage on the surface and hidden onset,there is currently a lack of effective screening and early diagnosis methods,leading to more than 70% of patients being in the advanced stage when they go to hospitals,but the treatment effect and prognosis of patients are poor,with the 5-year survival rate lower than 45%.Therefore,it is urgently to search for effective molecular markers and develop relevant targeted drugs to improve the prognosis of OC patients.Chemokines are small molecular proteins that interact with seven-transmembrane G-protein-coupled receptors and play a key role in inducing chemotaxis,promoting differentiation,leukocytosis and causing tissue exosmosis.C-X-C motif receptor 3(CXCR3)is a member of the chemokine receptor family,and its increased expression promotes recruitment of Tumor infiltrating lymphocytes(TILs).It plays an important role in the migration and localization of TILs.Among them,Chemokine C-X-C motif ligand 9(CXCL9)and Chemokine 10(CXCL10)produced in tumor tissue bind to CXCR3,CXCR3+T cells are mediated to migrate to the tumor site and play an important role in the attack and clearance of tumor cells.Recent studies have found that high expression of CXCL9 and CXCL10 in cutaneous melanoma,colorectal cancer,breast cancer and other malignant tumors is associated with a good prognosis,but the expression and clinical significance of CXCL9 and CXCL10 in EOC remain unclearly.Therefore,this study aims to provide a new theoretical basis for targeted therapy of OC by exploring the role of CXCL9 and CXCL10 in EOC and their relationship with clinicopathology.Method:1.GEPIA database was used to analyze the expression of CXCL9 and CXCL10 in OC and their relationship with the prognosis of patients.Meanwhile,the correlation between CXCL9 and CXCL10 expression levels in OC was analyzed using the TIMER database.2.The expression levels of CXCL9 and CXCL10 in 43 cases of EOC tissue and 15 cases of ovarian benign tumor were detected by immunohistochemical staining.3.Statistical methods: SPSS 23.0 software was used for comparative analysis of the data obtained in this experiment.Counting data is expressed in terms of cases or percentage(%).Chi-square test was used to analyze the differences in the expression of CXCL9 and CXCL10 between the EOC group and the benign lesion group,as well as the differences in the positive expression of CXCL9 and CXCL10 in EOC and the clinicopathological features.Spearman rank correlation test was used to analyze the correlation between CXCL9 and CXCL10 expression levels in EOC.P<0.05 was considered statistically significant.Research Result:1.The GEPIA database found that the expression levels of CXCL9 and CXCL10 in OC were significantly higher than those in normal tissues.High expressions of CXCL9 and CXCL10 were associated with better prognosis in OC patients(P<0.05).In addition,there was a positive correlation between the expression levels of CXCL9 and CXCL10 in OC through the analysis of the TIMER database(cor=0.69,P<0.05).2.Immunohistochemical staining showed that CXCL9 was highly expressed in EOCs compared with normal tissues,and had significant differences in different degrees of differentiation(P<0.05).CXCL10 was highly expressed in EOCs and had significant differences in tumor size and differentiation degree(P<0.05).3.There was a positive correlation between the expression levels of CXCL9 and CXCL10 in EOC(r=0.407,P=0.007).Conclusion:1.CXCL9 and CXCL10 are highly expressed in OC,and positive expressions of CXCL9 and CXCL10 are associated with better prognosis in OC patients.In addition,the expression levels of CXCL9 and CXCL10 in OC are positively correlated.2.Immunohistochemical staining showed that CXCL9 was highly expressed in EOCs and had significant differences in different degrees of differentiation.CXCL10 was highly expressed in EOCs and varied significantly in tumor size and differentiation.3.Immunohistochemical staining showed that the expression levels of CXCL9 and CXCL10 in EOC were positively correlated.
