| Research methods: 1.Public Clinical Databases: KEGG database,GEPIA database and Kaplan-meier Plotter Database.2.Clinical specimens: A: Case data of cases diagnosed with CCRCC in parallel surgery at the Affiliated Hospital of Qingdao University and their paraffin specimens were collected in 147 cases for immunohistochemical staining.B: Fresh tissue specimens diagnosed with CCRCC in surgery at the Affiliated Hospital of Qingdao University were collected in 30 cases for PCR detection.3.Methods: In this study,we used bioinformatics analysis,PCR and immunohistochemistryResearch background: Clear cell renal cell carcinoma(CCRCC)is the most common type of primary malignancy in adults with specific histological features of the kidney.Current studies suggest that CCRCC is a metabolic disease and metabolic reprogramming plays an important role in the development of CCRCC.In this research,we examined the expression of sterol regulatory element binding protein(SREBP1),fatty acid synthase(FASN)and carnitine palmitoyl transferase1A(CPT1A)and 2(CPT2)in CCRCC to preliminarily explore the abnormal alterations of lipid metabolism-related factors during the development of CCRCC,and the relationship between them and biological behaviors,and to provide a target for CCRCC.Results: 1.The expression of SREBP1 and FASN in CCRCC was higher than that in adjacent tissues(p<0.05).2.The expression of CPT1 A and CPT2 in CCRCC was lower than that in adjacent tissues(p<0.05).3.The expression of CPT1 A and CPT2 was higher in high-grade CCRCC than in low-grade CCRCC(p < 0.05).4.The expression of CPT1 A in CCRCC was correlated with clinical stage and sarcomatoid differentiation(χ2=4.196,4.556,p<0.05).The expression of CPT2 was correlated with clinical stage(χ2=4.286,p<0.05).5.Kaplan-Meier survival analysis and one-way Cox regression analysis showed that the expression of CPT1 A and CPT2 correlated with the overall survival time of CCRCC patients.Conclusion: SREBP1,FASN,CPT1 A and CPT2 may be involved in the occurrence,progression and lipid metabolism of CCRCC.The expression levels of CPT1 A and CPT2 are related to the WHO/ISUP classification and clinical stage of CCRCC,and can be used as indicators to determine the prognosis of CCRCC. |
