| Objective: The role of fibrinogen-like protein 1(FGL1)in tumor immunity has been reported before.However,the role of FGL1 in prostate cancer(PC)remains unclear.This study aimed to investigate the effect of FGL1 on apoptosis of prostate cancer cell.Methods: Data was downloaded from The Cancer Genome Atlas(TCGA)database,and differentially expressed genes(DEGs)were identified.We performed univariate analysis to confirm a gene set related to the overall survival(OS)among DEGs.The diagnostic value of the genes was evaluated using Receiver operating characteristic(ROC)curve.The single-gene difference analysis of FGL1 in prostate cancer was performed via R language.The bioinformatics determined the correlation between the expression levels of FGL1 and DNAJC12 in prostate cancer.The expression of FGL1 was silenced by si RNA in PC3 prostate cancer cell.And further the level of apoptosis was detected by Flow cytometry.Cell proliferation was detected by KI67.The expression of DNAJC12,Bcl-2and cleaved-caspase-3 was detected by Western blot.Results: By analyzing the difference of prostate cancer data downloaded from TCGA database,a total of 3011 DEGs,1254 up-regulated genes and 1757 down-regulated genes were obtained.Five target genes(RRM2,SLPI,FGL1,UGT2B4,FCGBP)were obtained by univariate analysis.Up-regulated gene FGL1 was identified as the selected gene in this study.The single-gene difference analysis showed that FGL1 had the highest positiverelation with DNAJC12.The OS of PC patients with high expression of FGL1 was significantly shorter than those with low expression of FGL1.We established efficient si RNA silences the expression of FGL1 in PC3 cell line.The level of apoptosis was increased,and the expression of cleaved-caspase-3 was increased,while the expression of DNAJC12 and Bcl-2 was reduced in FGL1 silencing prostate cancer cells.In addition,the results of Flow cytometry and KI67 suggest FGL1 silencing inhibited proliferation and promoted apoptosis of prostate cancer cell.Conclusions: Inhibition of FGL1 promotes the apoptosis of prostate cancer cell.FGL1 may be an independent prognostic marker and therapeutic target in PC. |
