Objective:The study investigated the differential expression of P62 in OSCC tumor tissues and adjacent non-tumor tissues,examining the changes of P62 in OSCC tumor tissues before and after preoperative induction chemotherapy with TP regimen,as well as the changes in tumor tissues after preoperative induction chemotherapy with TP regimen combined with nimotuzumab.Furthermore,this study analyzed the impact of adding nimotuzumab to preoperative induction chemotherapy on the expression of P62.Methods:From September 2020 to July 2022,oral and maxillofacial surgery cases at the First Hospital of Shanxi Medical University diagnosed with OSCC were collected.A total of 20 OSCC patients who did not receive preoperative induction chemotherapy were selected,and their tumor tissue samples and adjacent non-tumor tissue samples were obtained.Afterwards,the expression of P62 protein in OSCC and non-tumor tissues was detected using immunohistochemistry(IHC)and real-time fluorescence quantitative polymerase chain reaction(RT-q PCR).Additionally,33 patients with locally advanced-stage OSCC were selected and divided into two groups under different treatment protocols,i.e.,the TP group(docetaxel and cisplatin)for preoperative induction chemotherapy,and the N-TP group(nimotuzumab combined with TP)for preoperative induction chemotherapy.Besides,tumor tissue samples were obtained by pre-treatment biopsy and post-treatment surgical resection in both groups.The expression changes of P62 in the two groups before and after medication were detected using IHC and RT-q PCR.All data were analyzed using SPSS 26.0 statistical software.The chi-square test was used for categorical data,and the mean ± standard deviation was used to represent continuous data.Between-group comparisons were performed using the t-test or rank-sum test.A significance level of P < 0.05 was considered statistically significant.Results:1.Expression of P62 in OSCC:IHC showed that the positive rate of P62 protein in OSCC tumor tissues was significantly higher compared to adjacent non-tumor tissues(P < 0.001).Besides,RT-q PCR revealed that the relative expression level of P62 m RNA in OSCC tissues was significantly higher than that in adjacent non-tumor tissues(P < 0.001).2.Effects of TP and N-TP regimens on P62 expression in locally advanced-stage OSCC:The results demonstrated that both the TP and N-TP groups showed a decrease in the average optical density values of P62 protein and the expression level of P62 m RNA in tumor tissues after medication,with statistically significant differences before and after treatment(P < 0.05).Also,there was no statistically significant difference in the change of average optical density values of P62 between the TP and N-TP groups before and after medication(P > 0.05).Similarly,there was no statistically significant difference in the change of P62 m RNA between the TP and N-TP groups before and after medication(P > 0.05).Conclusion:1.P62 is highly expressive in OSCC.2.Preoperative induction chemotherapy group with TP or N-TP allows for downregulation of the P62 expression in locally advanced OSCC.3.Adding nimotuzumab to TP preoperative induction chemotherapy may not further decrease P62 expression.
