Objective:To determine whether β-catenin,Smo and Cx43 were co-expressed in the second heart field and the endoderm,we observed the expression patterns of these proteins.To explore whether Cx43 interacts with signaling molecules of the Wnt/β-catenin pathway or Shh pathway in the development of the second heart field Methods:Serial paraffin sections of the mouse embryos at Embryonic day(E)10-E12 were selected for immunohistochemical staining,HE staining and immunofluorescence staining.The primitive gut of mouse embryos at E11 was separated for WB and Co-IP.Results:1.The co-expression of Cx43 and Isl1 was observed in the cells of pharyngeal mesenchyme and dorsal wall of the pericardial cavity of mouse embryos at E10-E12,Isl1 positive cells increased while Cx43 positive cells increased.2.At E10.5-E11,Cx43 was co-expressed with β-catenin in some cells in the Isl1-positive conical region located in the ventral side of the foregut and in the epithelial cell cord extending from the cells in the ventral part of the foregut endoderm.Co-expression of Cx43 with Smo was found in the foregut endoderm and the epithelial cell cord extending from the ventral of the foregut.3.Co-IP results indicate that there is an interaction between Cx43 and β-catenin,but Cx43 has no interaction with Smo.Conclusion:Cx43,wnt pathway,and Shh pathway are all involved in the development of the second heart field;Cx43 interacts with β-catenin to participate in the development of the second heart field.While Cx43 does not interact with Smo.
