ObjectivePancreatic cancer is one of the worst malignant tumors in the world.Hidden onset,rapid progress and early metastasis are the early clinical characteristics of pancreatic cancer.Because there are no typical clinical symptoms,the disease has entered a late stage when the disease is diagnosed.At this time,patients often have lost the opportunity of surgery and missed the best treatment period.Due to the biological and clinical characteristics of pancreatic cancer,it is difficult to make early diagnosis.At present,the diagnosis of most pancreatic cancer mainly depends on the clinical manifestations,characteristics and corresponding impact test results,However,it is difficult for traditional diagnosis and treatment measures to effectively carry out early screening for pancreatic cancer.Therefore,more sensitive and specific diagnostic techniques and indicators are needed to improve the probability of early diagnosis and treatment of diseases.Transmembrane 4 superfamily(TM4SF),It is a small molecule transmembrane protein that can combine with cell surface molecules such as growth factor receptor,immunoglobulin superfamily and integrin to form a four transmembrane protein network,thus regulating cell signal transduction and mediating cell proliferation,differentiation,adhesion,migration and other physiological processes.CD81 is an important member of TM4 SF,which is widely expressed in breast,lung,prostate,brain and lymph nodes,and may become a potential tumor treatment target.In this study,we will focus on the role of CD81 in pancreatic carcinogenesis and explore its expression and effects on pancreatic cancer cell proliferation,invasion,migration,apoptosis.This study will provide new therapeutic strategies and research ideas for pancreatic cancer progression,metastasis inhibition,and patient prognosis improvement.MethodsThrough bioinformatics analysis,the differential expression of CD81 in pancreatic cancer and pan cancer was analyzed using R language and online databases such as GEPIA2,The expression levels of CD81 in cancer tissues and adjacent tissues were determined by fluorescence quantitative PCR(q RT-PCR)and WB.The expression of CD81 was detected in pancreatic cancer PANC-1 、 Bx PC-3 cells and h TERT-HPNE human normal pancreatic duct cells in vitro,CD81 protein amounts were quantified by Western Blot(WB).Pancreatic cancer data were downloaded from TCGA database,In R software,the prognosis of CD81 in pancreatic cancer patients was plotted by using the survival package.Amplification by plasmid transfection of CD81 in pancreatic cancer cells,CCK8 、Transwell 、scratch assay 、The cell apoptosis for proliferation,invasion,migration and apoptosis of pancreatic cancer cells.ResultIn pancreatic cancer tissues,the expression level of CD81 was significantly lower than that in adjacent noncancerous tissues.Within pancreatic cancer cells,the expression level of CD81 was significantly lower than that in normal pancreatic cancer cells.Lower intratumoral expression levels of CD81 in pancreatic cancer patients represent worse prognosis and shorter survival.By upregulating the expression levels of CD81,the proliferation,invasion and migration levels of pancreatic cancer cells were significantly inhibited,and the apoptotic rate was significantly higher.ConclusionCD81 can inhibit the proliferation,invasion and migration of pancreatic cancer cell,as well as induce the apoptosis of pancreatic cancer cell.
