Correlation Of TRPV1 Rs222747 Gene Polymorphism With Adverse Cardiovascular Events After Induction Of General Anesthesia In Surgery Patients | Posted on:2024-08-25 | Degree:Master | Type:Thesis | Country:China | Candidate:D Q Jing | Full Text:PDF | GTID:2544307148978579 | Subject:Anesthesiology | Abstract/Summary: | PDF Full Text Request | Objective:The high incidence of adverse cardiovascular event(ACVE)occurs after induction of anesthesia before the start of surgery without knowing the underlying mechanism.Transient receptor potential vanilloid-1(TRPV1)and its downstream effector peptide,calcitonin gene-related peptide(CGRP)and substance P(SP)play important roles in regulation of cardiovascular functions.Gene polymorphism has a certain impact on the functions of related proteins.The purpose of this study was to examine the association of TRPV1 rs222747 gene polymorphism with ACVE after induction of general anesthesia in surgery patients given the same dose of drugs,at the same rate of drug delivery during the anesthesia induction.Methods:Two hundred and thirty-six patients of both sexes,ASA physical grade I~Ⅲ,aged20~65 years,undergoing elective non-cardiovascular surgery under general anesthesia were enrolled and assigned to ACVE group(n=117)and non-ACVE(NACVE group,n=119)according to the occurrence or not of ACVEs after anesthesia induction.Blood samples were collected at 30 min before the intravenous anesthesia induction and at15 min after endotracheal intubation.Real-time fluorescence quantitative PCR was used to genotype TRPV1 rs222747,and the genotypes and alleles of TRPV1 rs222747 were analyzed.Enzyme-linked immunosorbent assay(ELISA)was employed to measure the serum concentrations of CGRP and SP before and after anesthesia induction.Results:TRPV1 rs222747 genotype GG ratio and allele G frequency were significantly higher in ACVE group(P < 0.05),with significantly lower levels of serum concentrations of CGRP and SP before and after anesthesia induction(P < 0.001),in comparison with NACVE group.The serum SP and CGRP concentrations of TRPV1rs222747 genotype GG were notably decreased before and after induction(P < 0.05).Logistic regression analysis indicated that increased age and TRPV1 rs222747 genotype GG were associated with increased incidence of ACVEs after induction of general anesthesia.Conclusion:The increased GG ratio and allele G frequency of TRPV1 rs222747 are associated with the decreased serum concentrations of CGRP and SP and the higher incidence of ACVEs after induction of general anesthesia.The underlying mechanisms warrant further investigation. | Keywords/Search Tags: | TRPV1, Gene polymorphism, Calcitonin gene-related peptide, Substance P, General anesthesia induction, Adverse cardiovascular events | PDF Full Text Request | Related items |
