Expression Of Hsa_circ_0000848 In Human Breast Cancer Serum And Its Effect On Proliferation And Migration Of Breast Cancer Cells | | Posted on:2024-04-21 | Degree:Master | Type:Thesis | | Country:China | Candidate:J Y Yang | Full Text:PDF | | GTID:2544307160490534 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Background and Objective: Breast Cancer(BC)is the most common cancer in women around the world.With the rapid development of comprehensive treatment of breast cancer,the 5-year survival rate is increasing gradually,but it is still a serious threat to women’s health.Different stages of breast cancer can lead to different prognosis,and early stage breast cancer has a good prognosis.It is important to improve the early diagnosis rate to reduce the morbidity and mortality.Circular RNAs are abundant,stable,tissue and stage specific non-coding RNAs that have become potential cancer targets.In order to improve the rate of early screening and individualized treatment of breast cancer,the aim of this study is to screen the differentially expressed circular RNA in breast cancer by gene sequencing and to explore its function,in order to find diagnostic markers and functional therapeutic targets of breast cancer.Methods:1.Preoperative serum samples,postoperative breast cancer and paracancerous tissue and patient clinical pathology data were collected from breast cancer patients who met the inclusion criteria in the third affiliated hospital of Guangzhou medical university.2.To screen the differentially expressed circ RNAs in the serum of three breast cancer patients and three normal people by gene sequencing.3.The technique of q RT-PCR was used to relative quantitative analysis the expression of hsa_circ_0000848 in the serum of patients with breast cancer and healthy people,as well as in the breast cancer and para-cancer tissues.Collect clinical pathology data of breast cancer patients who met the inclusion criteria.4.To interfere the expression of hsa_circ_0000848 by si RNA,and to explore the effect of interfering the expression of hsa_circ_0000848 on the proliferation and migration of MDA-MB-231 breast cancer cells by CCK8 and scratch test.Results: 1.Compared with healthy people,the expression of hsa_circ_0000848 in the serum of patients with breast cancer was significantly higher than that of the healthy persons(P < 0.0001).To assess the diagnostic value of hsa_circ_0000848 as a serum marker for breast cancer,the diagnostic ability of hsa_circ_0000848 was analyzed using a ROC curve with an area under the ROC curve of 0.877(95% CI,0.8256-0.928);the maximum value of the Youden index was 0.571;The sensitivity was 1 and the specificity was 0.571.2.By analyzing the relationship between the relative expression of hsa_circ_0000848 and the clinicopathological features of breast cancer,we found that Ki-67(P = 0.034)and PR(P = 0.013)were statistically correlated,the serum level of Ki-67 was higher in patients with high KI-67 expression and negative PR.Comparing the expression of hsa_circ_0000848 in the serum of early stage breast cancer patients and healthy people,hsa_circ_0000848 showed a high expression trend in the serum of early stage breast cancer patients(p < 0.0001),the AUC area was 0.906(95% CI,0.859-0.953),and the maximum value of Youden index was 0.628,the sensitivity was 1 and the specificity was 0.628.3.CCK8 and Scratch Test showed that hsa_circ_0000848 could inhibit the proliferation and migration of MDA-MB-231 cells.Conclusion: 1.Hsa_circ_0000848 was significantly up-regulated in serum of breast cancer(p < 0.0001),and its high expression was related to Ki-67 high expression and PR negative.2.Interfering with the expression of hsa_circ_0000848 in MDA-MB-231 breast cancer cells can inhibit the proliferation and migration of breast cancer cells.3.Hsa_circ_0000848 has an increasing tendency in serum expression of early breast cancer(p < 0.0001).Hsa_circ_0000848 has the potential to be a serum marker for the diagnosis of breast cancer,which may be beneficial to the early screening of breast cancer. | | Keywords/Search Tags: | breast cancer, biomarkers, circular RNA, proliferation, migration | PDF Full Text Request | Related items |
