| ObjectiveTo explore the back skin injury in mice to construct an animal model of Chinese cobra bite ulcer by use of Cytotoxin(CTX).The back wound healing induced by cytotoxin and the effect of anti-cobra venom on wound healing in mice were analyzed by group intervention.MethodsDifferent doses of Chinese cobra cytotoxin were used to induce dorsal skin damage in mice and the effect was observed.On the 3rd day after the injury model was successfully prepared,the wound was treated with anti-cobra venom(Anti group)or antivenom combined with methylprednisolone(MPS group)injected into the tail vein and no specific treatment were given(Control group),the wound changes and healing days were observed and recorded.Result1.The success rate of mouse model preparation using Chinese Cobra CTX was43.5%for 1.5 times MND,60.9%for 1.8 times MND,and 94.4%for 2 times MND,and there were statistical differences between the three groups.The failure rate of mouse model preparation using Chinese Cobra CTX was 56.5%for 1.5 times MND,39.1%for 1.8 times MND,and 5.6%for 2 times MND,and there was no statistical difference between the three groups.2.Anti-cobra venom and anti-cobra venom combined with hormone therapy were given intravenously on the 3rd day after the preparation of the animal model,and the wound area difference and healing rate of mice in the anti-cobra venom combined with hormone therapy group on the 6th day of modeling were statistically different from those in the control group.3.The healing days in the Control group were(15.9±2.3)days,the median healing days in the Anti group were 15(3)days,and the healing days in the MPS group were(15±2.4)days,with no statistical difference in the healing days between the three groups.Conclusion1.Higher success rate of mouse back ulcer model preparation by using 2 times dose of MND Chinese cobra CTX.2.CTX-induced back wounds in mice showed a greater reduction in wound area on the 3rd day after a single injection of anti-cobra venom combined with methylprednisolone treatment.3.Anti-cobra venom serum or anti-cobra venom serum combined with hormone single injection treatment did not significantly reduce the number of days of CTX-induced skin wound healing. |
