| Objective:(1)To establish an animal model of aortic valve calcification by giving high cholesterol diet and vitamin D2;(2)To investigate the expression of osteopontin,RUNX2 and CD44 in the anti-calcification effect of resveratrol on aortic valve;(3)To study whether resveratrol can improve the anti-calcification of aortic valve.Methods: Fifteen male New Zealand white rabbits were randomly divided into three groups: the control group with normal diet,the model group with 0.5% high cholesterol diet and vitamin D2,and the RES group with 0.5% high cholesterol diet and vitamin D2 and resveratrol.After 12 weeks,the rabbits were euthanized,peripheral blood was collected from the apex of the heart,and the heart was dissected to obtain aortic valve tissue.The supernatant was collected from peripheral blood after centrifugation,and the expressions of osteopontin(OPN),dwarf-related transcription factor 2(RUNX2)and CD44 were analyzed by enzyme-linked immunosorbent assay.At the same time,the concentration levels of calcium ion and alkaline phosphatase(ALP)in peripheral blood were determined,and the aortic valve was stained with HE to observe the calcification of the valve.Results: 1.A large number of blue-purple calcified particles can be observed on the HE stained sections of the arterial valve in the model group,which is obviously different from that observed in the control group,while the number of calcified particles on the HE stained sections in the RES group is between the two;2.Compared with the model group,the levels of osteopontin,RUNX2,serum calcium ion and alkaline phosphatase in RES group were significantly decreased(P < 0.05),but they were still higher than those in the control group.3.Compared with the expression of CD44 in peripheral blood of model group,the level of CD44 in RES group was significantly decreased(P <0.05).Conclusion:(1)It is feasible to establish an animal model of aortic valve calcification by feeding high cholesterol feed and vitamin D2 supplement;(2)Osteopontin,RUNX2 and CD44 are involved in the calcification of aortic valve;(3)Resveratrol can delay the progress of calcification of aortic valve by down-regulating the expression of osteopontin,RUNX2 and CD44. |
