Objective This study aims to solve the problem of poor stability of oily traditional Chinese medicine powder.The stability of the powder and its therapeutic effect are affected by the shortcomings of its volatile components,which are easy to be oxidized,volatile and loose.In this study,taking Lingzhu Powder as an example,Pickering emulsion technology was introduced to improve the stability of the volatile oil of Acorus tatarinowii in pediatric medicine Lingzhu Powder under the concept of"combination of medicine and supplement",so as to ensure the stability and safety of Lingzhu powder,and to provide reference value and scientific basis for the improvement of stability of other oil-containing traditional Chinese medicine powder.Methods 1.Characterization of related properties of each decoction piece powder in Lingzhu Powder and preparation method of Pickering emulsion;The most suitable decoction piece powder was selected as the stabilizer for the preparation of Pickering emulsion.The concentration of stabilizer,the ratio of oil to water and the preparation method were determined by comparing the amount and state of emulsion formation.2.Investigation on Improving the Stability of Volatile Oil from Acorus tatarinowii in Lingzhu Powder by Pickering emulsion technology;Compare the retention rate,malondialdehyde(MDA),and peroxide(POV)content of Acorus tatarinowii oil in thermal environment,pH value,Fe3+,and AIBA in the original crude oil group,oil-water mixture group,and Pickering group.At the same time,screen the sensitive compounds of Acorus tatarinowii oil in different environments,and analyze the intragroup differences and corresponding changes of the sensitive compounds.3.Investigation of powder and pharmaceutical properties of Lingzhu Powder before and after improvement;In order to clarify the impact of Pickering emulsion technology on the properties of Lingzhu powder,the Acorus tatarinowii oil introduced into Pickering emulsion technology was returned to the entire formula,and its moisture,particle size,appearance uniformity,color difference value,bulk density,contact angle,angle of repose,etc.were investigated.Influence factor experiments,accelerated experiments,and long-term experiments were conducted to analyze the differences in powder morphology,retention rate,and change trend of sensitive compounds before and after improvement,To clarify the effect of Pickering emulsion technology on the stability of Acorus tatarinowii oil.Results 1.Characterization of related properties of each decoction piece powder in Lingzhu Powder and preparation method of Pickering emulsion;By investigating the solubility,microstructure,contact angle,angle of repose,particle size,and type of formation of various pieces of powder,it was clear that pearl powder was used as a stabilizer for the preparation of Pickering emulsion.Based on the amount and state of formation,the optimal conditions for the formation of Pickering emulsion were determined as follows:pearl powder concentration 0.065 g·m L-1,oil-water ratio 9:11(V1:V2),and the preparation method was high-pressure homogenization(500 bar,3 times,20 min,normal temperature).2.Investigation on Improving the Stability of Volatile Oil from Acorus tatarinowii in Lingzhu Powder by Pickering emulsion technology;Compared with the crude oil group,the retention rate of the Pickering group significantly increased under the thermal environment,pH value,Fe3+,and AIBA,while the MDA and POV values significantly decreased.The trend of changes in sensitive compounds selected through analysis indicates that after the introduction of Pickering emulsion technology,the Pickering group has a slowing effect on the changes in sensitive compounds,and even has a reversal effect on some compounds.In addition,compared to the crude oil group,the standard deviation of the Pickering group is also smaller,further proving that it has a strong stabilizing effect on Acorus tatarinowii oil from the perspective of chemical composition changes.3.Investigation of powder and pharmaceutical properties of Lingzhu Powder before and after improvement;The particle size,water content,and appearance uniformity of the improved Lingzhu Powder comply with the provisions of the Chinese Pharmacopoeia and have no statistical differences from the original Lingzhu Powder.Its color difference value,contact angle,angle of repose,and bulk density are also consistent with the original Lingzhu Powder.The results of near-infrared and mid-infrared spectroscopy also indicate that the introduction of Pickering emulsion technology has not destroyed the original components in Lingzhu Powder,nor has it generated new chemical components.In influencing factor experiments,accelerated experiments,and long-term experiments,the retention rate of Acorus tatarinowii oil and the changes in sensitive compounds contained in Lingzhu Powder before and after improvement were analyzed,indicating that the introduction of Pickering emulsion technology significantly improved the retention rate of Acorus tatarinowii oil,and the change range of sensitive compounds was slowed.The above research shows that the introduction of Pickering emulsion technology has improved the stability of Acorus tatarinowii oil in Lingzhu Powder,thereby ensuring the stability of the preparation.Conclusion The introduction of Pickering emulsion technology under the concept of"Combination of medicine and adjuvant"can be used to improve the stability of volatile components in oily traditional Chinese medicine powders,providing a reference basis for improving the stability of other oily traditional Chinese medicine powders.
