Purpose: To investigate the expression levels of DNMT3 A and NLRP3 in peripheral venous blood of patients with chronic heart failure and to detect the expression of DNMT3 A and NLRP3 in the myocardial tissue of mice with knockout NLRP3 genotype and wild-type mice in animal experiments,and to investigate the expression of DNMT3 A and NLRP3 in chronic heart failure and their correlation.Methods: A total of 40 patients with chronic heart failure who visited a tertiary hospital in Hangzhou from May 2022 to December 2022 were selected and divided into HFr EF group,HFmr EF group and HFp EF group according to the guidelines for the diagnosis and treatment of chronic heart failure.The expression levels of DNMT3 A and NLRP3 in peripheral venous blood were analyzed.In terms of animal experiments,male 8-week-old C57BL/6 mice were randomly divided into heart failure group of wild mice(group A,n=8)and wild mouse control group(group B,n=7);Male 8-week-old NLRP3-out(NIRP3-/-)mice were randomly divided into NLRP3-/-mouse heart failure group(C group,n=7),NLRP3-/-mouse control group(D group,n=6).Groups A and C were induced to construct a chronic heart failure model by intraperitoneal injection of isoprenaline for 2 weeks,and the surviving mice in each group were perfected by cardiac ultrasonography,cardiac tissue was taken after dissection,and DNMT3 A content level and influencing factors were analyzed by enzyme-linked immunosorbent assay.Results: The content of peripheral plasma DNMT3 A in HFr EF patients was significantly higher than that in HFmr EF group,HFp EF group and control group(p<0.05).With the exacerbation of chronic heart failure,the expression level of NLRP3 in peripheral venous blood plasma gradually increased,and the difference was statistically significant in HFr EF group>HFmr EF group>HFp EF group(16.07±2.71 vs.13.56±1.28 vs.11.78±1.52)(p<0.05).In animal experiments,the content of DNMT3 A in mouse heart tissues was determined as(1)A>C(1.51±0.15 vs.1.10±0.18,p<0.05),the difference was statistically significant,while the difference between group B and group D was not statistically significant(p>0.05);(2)A>B(1.51±0.15 vs.1.05±0.07,p<0.05),the difference was statistically significant,while the difference between group C and group D was not statistically significant(p>0.05).Conclusion: Through the analysis of clinical research results,it is believed that DNMT3 A and NLRP3 expression are increased in patients with chronic heart failure,and NLRP3 can reflect the severity of chronic heart failure to a certain extent.The analysis of animal results further confirmed that DNMT3 A and heart failure severity were positively correlated,and it was found that lower levels of DNMT3 A content may be related to NLRP3 gene deletion.
