| Part 1 Effects of TGF-β1 on biological properties and cytokine expression of human periodontal ligament fibroblasts in inflammationObjective:To investigate the effects of Transforming growth factor β1(TGF-β1)on the biological properties of human periodontal ligament fibroblasts(hPDLFs)stimulated by Lipopolysaccharide from Porphyromonas gingivalis(Pg-LPS)and the expression of inflammatory cytokines.Methods:hPDLFs were obtained and identified by immunohistochemistry,The stimulating concentration of Pg-LPS was determined by qRT-PCR and CCK-8.The hPDLFs were divided into blank control group and 1,10,100 ng/mL TGF-β1 groups,cell proliferation was measured by CCK-8 at 72 hours,cell migration was measured by scratch test and Transwell Chamber Test at 24 hours,cell cycle of hPDLFs was measured by Flow cytometry test at 72 hours.The expression of Forkhead/winged helix transcriptional factor p3(Foxp3),Interleukin-6(IL-6)and Epstein-Barrvirus-induced gene 3(EBI3)mRNA in hPDLFs at 72 hours was detected by qRT-PCR,and the expression of Foxp3,IL-6 and EBI3 proteins in hPDLFs at 72 hours was detected by Western blot.Results:The results of immunohistochemistry showed that anti-vimentin was positive and anti-keratin was negative;At the concentration of 100 μg/mL Pg-LPS,the expression of IL-6 mRNA in hPDLFs was increased(P<0.05)and the proliferation of hPDLFs was decreased(P<0.05).TGF-β1 at 10 ng/mL and 100 ng/mL could increase the proliferation of hPDLFs stimulated by 100 μg/mL Pg-LPS(P<0.05),TGF-β1 could promote the migration of hPDLFs stimulated by 100 μg/mL Pg-LPS(P<0.05)1,10,100 ng/mL TGF-β1 could accelerate the cell cycle of hPDLFs stimulated by 100 μg/mL Pg-LPS(P<0.05),1,10,100 ng/mL TGF-β1 could inhibit the expression of IL-6 gene and protein(P<0.05);1,10 ng/mL TGF-β1 could increase the expression of EBI3 gene and protein(P<0.05),1.10 ng/mL TGF-β1 could increase the expression of Foxp3 gene and 10 ng/mL TGF-β1 could increase the expression of FOXP3 protein(P<0.05).Conclusion:TGF-β1 can promote the proliferation and migration of hPDLFs in inflammation condition,up-regulate EBI3 and inhibit inflammation,which may be related to the expression of transcription factor Foxp3 and reveal a new mechanism of inflammation regulation.Part 2 Case presentation:Digital application of periodontal sequence therapyObjective:To observe the effect of digital application on periodontal sequence therapy.Methods:Five patients with stage Ⅲ/Ⅳ periodontitis were selected and diagnosed in the periodontal disease treatment center of our hospital.According to the different periodontal lesions of patients,periodontal endoscopy was used for periodontal basic treatment.According to the pathological changes of root bifurcation in the posterior tooth region,the corresponding surgery was adopted;According to the looseness of the anterior tooth area,digital oral scanning design and 3D printing periodontal splint were adopted for fixation.During the periodontal treatment of patients,regular follow-up and re-examination were carried out to record the patients’oral photos before and after treatment,the full oral periodontal exploration record form and the apical film to evaluate the effect of periodontal treatment.Results:After the basic treatment under the periodontal endoscope,the periodontitis of 5 patients was significantly improved compared with that before the treatment,and the sites with the depth of periodontal exploration more than 6 mm had good therapeutic effect,which could avoid periodontal surgery to a certain extent,but the third degree sites of root bifurcation lesions in the posterior teeth still needed surgical treatment.The anterior tooth looseness can be effectively fixed by 3D printing periodontal splint on the first degree affected teeth without discomfort;The loose anterior teeth can be effectively retained by intentional replantation and 3D printing periodontal splint fixation of the teeth with more than Ⅱ degree of looseness and combined periodontal and pulp lesions.Conclusion:All kinds of digital applications in periodontal sequential treatment can improve the efficacy of periodontal treatment and reduce the discomfort of patients,and provide a new treatment plan for more effective retention of natural teeth. |
