The Relationship Between CircRNA-001846 And Invasion And Metastasis Of Breast Cancer And Its Clinical Value

Objective:Breast cancer is one of the most common malignant tumors in women.Breast cancer metastasis can seriously reduce patients’ quality of surviving,and even lead to death.Based on our previous studies,circ RNA-001846 was found to be highly expressed in breast cancer tissues,but its relationship with breast cancer and its clinical value remain unclear.This study aims to explore the relationship between circ RNA-001846 and the breast cancer,to determine whether circr NA-001846 can be used as a serological indicator for the diagnosis of breast cancer or to predict the metastasis of breast cancer.Methords: Absolute quantitative PCR was used to detect the expression of circ RNA-001846 in serum of 50 breast cancer patients and 50 healthy women.Receiver operating characteristic(ROC)curve was used to determine the diagnostic value of circ RNA-001846 in breast cancer.The expression of circ RNA-001846 in 31 pairs of breast cancer tissues and adjacent tissues was detected,and the correlation of clinicopathological features was analyzed.The relationship between the expression level of circ RNA-001846 and the proliferation of breast cancer cells was analyzed by CCK-8 assay and plate cloning assay.Wound healing assay and Transwell invasion and migration assay were used to investigate the relationship between the expression level of circ RNA-001846 and the invasion and migration ability of breast cancer cells.Results : Compared with healthy women,the expression of circ RNA-001846 was upregulated in the serum of breast cancer patients.The ROC curve showed that circ RNA-001846 had a sensitivity of 86% and a specificity of 70% as a serum marker for the diagnosis of breast cancer,which had a good diagnostic efficacy.The expression of circ RNA-001846 in breast cancer tissues was higher than that in adjacent tissues.The correlation analysis of clinicopathological features showed that the expression of circ RNA-001846 was related to lymph node metastasis.By comparing the expression of circr NA-001846 in human breast epithelial cells,non-triple-negative breast cancer cells and triple-negative breast cancer cells,we found that the expression of circ RNA in breast cancer cells was higher than that in human breast epithelial cells,and the expression of circ RNA in triple-negative breast cancer cells was higher than that in non-triple-negative cells.After the expression of circ RNA-001846 was inhibited,the proliferation,invasion and migration of breast cells were decreased.Conclusion:circ RNA-001846 is positively correlated with the proliferation,invasion and migration of breast cancer.The expression level of circ RNA-001846 has good sensitivity and specificity for the diagnosis of breast cancer,and may also be used to predict the metastasis of breast cancer patients.
