| Background and purposePreeclampsia(PE)is a pregnancy-specific phenomenon,mainly characterized by high blood pressure and albuminuria after 20 weeks of pregnancy,and is one of the major cause of morbidity and mortality in mother-to-be and the unborn child.Fresh reviewing have shown that circr RNA play a pivotal role in the progression of preeclampsia.The analysis aimed to quantitatively and differentially express cir-104700、miR-141-5p and MAPK1 in preeclampsia and normal placenta,so as to explore the engagement or inordinately exhibited cirRNAs in the pathophysiology of preeclampsia through MAPK signal transduction pathway.Methods36 mother-to-be who experienced caesarian section were contained in this study,including 18 patients with preeclampsia and 18 ordinary pregnant women.Preferentially expressed cirRNA in placenta were obtained through literature review,The cir-104700 with significantly differential overexpression was selected,meanwhile related miRNAs were predicted.GO enrichment and KEGG signaling pathway were used to analyze the cell signaling pathways that miRNA might participate in.It was found that MAPK signaling channels may be involved in their cellular signaling channel function,meanwhile,the key gene MAPK1 of MAPK pathway with targeted binding sites with miR-141-5p(miRNA predicted to target cir-104700)was selected.Real-time PCR was used to detect the mRNA expressions of cir-104700,miR-141-5p and MAPK1 in preeclampsia and normal placental tissues,and Co-expression levels between the pairs were also analyzed.ResultsAnalysis of the GSE84260 dataset yielded a low expression trend for the manifestation of miR-141-5p.GO and KEGG pathway flourishing analysis was accomplished on genes from miR-141-5p.KEGG analysis explained that the aimed gene pathways convoluted in up-regulated genes may include calcium signaling pathway,phosphatidylinositol signaling system,stress-activated-protein-kinase(SAPK)pathway,etc.The binding site of miR-141-5p to MAPK1 was also predicted.Matchmaking of the clinical statistics between the two groups pointed that age,BMI,urinary protein,systolic,and diastolic BP were notability superior in group PE than those in group NO [age:(32.72 ± 5.988),(27.89 ± 3.998)(t=2.848,p=0.007);The BMI are:(30.938 ± 4.341)and(26.63 ± 2.833)(t=3.526,respectively,p=0.001);Urinary proteins are:(2.44 ± 1.042)and(0.06 ± 0.236)(t=9.490,respectively,P<0.001);The systolic blood pressure was respectively:(156.89 ±9.773)and(109.17 ± 9.038)(t=15.210),P<0.001);Diastolic blood pressure is:(103.11 ± 6.816)and(78.28 ± 7.419)(t=10.458 respectively,P<0.001)].The neonate weight in group PE was notability lower than in group NO [neonatal birth weight was:(2175.00 ± 808.093)and(3450.00 ± 264.575)](t=-6.362,P <0.001).The differences in the manifestation levels of target genes detected by qRT-PCR suggested that the expression status of circ-104700 in placental tissues of group PE was up-guided(P<0.05),the expression of mir-141-5p in placental tissues was down-guided(P<0.05),while the expression of MAPK1 mRNA was up-guided(P<0.05).Positive correlation between cir-104700 and MAPK1 mRNA(r=0.54,P<0.001);positive correlation between miR-141-5p and MAPK1 mRNA(r=-0.36,P=0.025).Conclusions1、Bioinformatic analysis proposes that miR-141-5p was poorly expressed in group PE and was significantly enriched in the SAPK signaling pathway,in addition,MAPK1 is the aimed bandaging spot of miR-141-5p.2、Age,BMI,urinary protein,systolic BP,and diastolic BP were significantly higher in group PE than in group NO,but the neonatal weight was significantly lower.3、The cir-104700 is up-regulated in placental tissue of patients with preeclampsia,suggesting that cir-104700 may be associated with the occurrence and development of preeclampsia.The manifestation levels of miR-141-5p decreased and the manifestation levels of MAPK1 mRNA increased in the placental flesh of pregnant women with preeclampsia.4、Positive correlation between cir-104700 and MAPK1 mRNA;positive correlation between miR-141-5p and MAPK1 mRNA. |
