Objective: To detect the expression of miR-145-5p in basal cell carcinoma(BCC)tissues and cells.Transfection of agomiR-145-5p&antagomiR-145-5p was used to observe the effects of on proliferation,migration invasion and EMT of TE 354.T cells.To observe the effect of agomiR-145-5p on the growth of BCC in nude mice.Methods: The expression of miR-145-5p in basal cell carcinoma tissues and cells was detected by RT-qPCR.Lipo2000 was used to transfect agomiR-145-5p&antagomiR-145-5p to TE354.T cells line.Cell viability was detected by CCK-8 kit and clone formation experiment.The migration and invasion of TE 354.T cells were detected by scratch test and Transwell assay.The expression of epithelial-mesenchymal transformation(EMT)-related proteins in TE 354.T cells was detected by Western blotting.The tumor model of nude mice was established by subcutaneous tumor formation,and was treated with agomiR-145-5p.Results: miR-145-5p expression was significantly decreased in BCC tissues and cells.After the overexpression of miR-145-5p,cell viability,the migration and invasion ability of TE 354.T cells decreased,and the ability of EMT decreased.The results after the inhibition of miR-145-5p were contrary.In vivo experiments,agomiR-145-5p can inhibit tumor proliferation.Conclusion: The present study found that the expression of miR-145-5p in BCC clinical tissues and cells was significantly decreased,and agomiR-145-5p could significantly inhibit cell viability,the proliferation,migration and invasion and EMT of TE 354.T cells.At the same time,agomiR-145-5p was also found to inhibit the proliferation of tumors in vivo,indicating that miR-145-5p plays an anticancer role,further exploring the pathogenesis of BCC,and providing a new effective target and theoretical basis for its gene therapy.
