| Objects:To detect the expression of cyclin dependent protein kinase9(CDK9)and inhibitor of apoptosis-stimulating protein of p53(iASPP)in normal cervical tissue,cervical squamous intraepithelial lesion and cervical cancer.Combined with the clinical data of patients,to analyze the relationship between the expression of CDK9 and iASPP and the clinicopathological features of cervical cancer was analyzed.To explore the role and mechanism of CDK9 and iASPP in the carcinogenesis of cervical cancer,and to further explore the role of CDK9 and iASPP in early diagnosis and clinical prognosis of cervical cancer.Methods:145 patients diagnosed by operation or biopsy in the first affiliated Hospital of Guangdong Pharmaceutical University and Boai Hospital of Zhong shan from September 2016 to 2020 were selected,there were 45 cases of cervical cancer(cancer group),40 cases of high-grade cervical squamous intraepithelial lesion(high-grade group),39 cases of low-grade cervical squamous intraepithelial lesion(low-grade group)and 21 cases of normal cervical tissue(control group).Immunohistochemical method was used to detect the expression of CDK9 and iASPP in cervical tissues of each group.The experimental data were statistically processed by SPSS26.0,and the mean between groups were compared by one-way ANOVA or Mann-Whitney U rank sum test;the rate between groups was compared by chi-square test;the relationship between the expression of CDK9 and iASPP and the clinicopathological features of cervical cancer was detected by chi-square test;the correlation between the expression of CDK9and iASPP in cervical cancer was analyzed by Spearman rank correlation coefficient.P<0.05 means the difference is statistically significant.Results:1.There was no significant difference in age,BMI,age of first sexual life,age of first pregnancy,times of delivery,times of abortion,smoking,menopause and other basic clinical data among the four groups(P>0.05).2.The positive expression rates of CDK9 in control group,low-grade group,high-grade group and cancer group were 19.4%,43.6%,55.0%and 68.9%,respectively,and the difference was statistically significant(X~2=15.46,P=0.001).3.In cervical cancer,the expression of CDK9 was related to clinical stage,depth of invasion and lymph node metastasis(P<0.05),but not to pathological type,degree of differentiation and vascular invasion(P>0.05).4.The positive expression rates of iASPP in control group,low-grade group,high-grade group and cancer group were 28.6%,51.3%,65.0%and 73.3%,respectively,and the difference was statistically significant(X~2=13.37,P=0.004).5.In cervical cancer,the expression of iASPP was related to clinical stage,depth of invasion and lymph node metastasis(P<0.05),but not to pathological type,degree of differentiation and vascular invasion(P>0.05).6.In the cervical cancer tissues with positive expression of CDK9,the positive expression rate of iASPP was higher,and the difference was statistically significant(X2=4.059,P=0.044).There was a positive correlation between CDK9 and iASPP expression in cancer group(r=0.355,P=0.017).Conclusion:1.With the aggravation of cervical lesions,the positive expression rates of CDK9 and iASPP gradually increased.When the positive expression of CDK9 and iASPP increases,the risk of cervical cancer increases.2.CDK9 and iASPP may be molecular markers for early diagnosis of cervical cancer,and may also be used as indicators for clinical stage,depth of invasion and lymph node metastasis of cervical cancer.3.CDK9 and iASPP have synergistic effect on the occurrence and progress of cervical cancer,and jointly promote the occurrence,development,invasion and metastasis of cervical cancer.CDK9 may act through iASPP molecular pathway.4.CDK9 and iASPP are expected to be potential drug targets for the treatment of cervical cancer and monitoring indexes for clinical evaluation of prognosis in patients with cervical cancer. |
