| Objective: Xuanwei lung cancer is a type of lung cancer with high incidence in Xuanwei area of Yunnan Province.To analyze the expression level of miR-551b-3p in tissue samples(lung cancer tissues and para-carcinoma lung tissues)and serum samples(preoperative and postoperative)of lung cancer patients in Xuanwei area,Yunnan Province,and analyze its correlation with clinic pathological features,then undertake cell biological function experimental study of lung cancer to learn about the relationship between miR-551b-3p and the development of lung cancer.In order to explore the clinical value and biological effect of miR-551b-3p in lung cancer patients of Xuanwei area,Yunnan Province.Methods: In this clinical study,52 lung cancer patients of Xuanwei area,Yunnan Province were included,RT-q PCR was used to detect the expression level of miR-551b-3p in lung cancer tissues,para-carcinoma lung tissues,preoperative and postoperative serum samples,after that,analyze the expression of miR-551b-3p in cancer tissues and normal tissues.Meanwhile,analyze the correlation between the expression of miR-551b-3p and clinic pathological characteristics,such as age,gender,smoking status,family history,tumor size,lymph node metastasis and stage.Lentivirus vector was used to transfer miR-551b-3p Gene,and to transfect on A549 cells and JT cells,miR-551b-3p over-expression cell group(miR-551b-3p mimics),negative control cell group(NC)and normal control cell group(Control)were set for A549 cells and JT cells respectively.To detect the transfection efficiency by RT-q PCR.To up-regulate the expression of miR-551b-3p in A549 cells and JT cells with lentiviral vectors,CCK-8 experiment,plate clone formation experiment,cell scratch test,Transwell chamber migration assay,Transwell chamber invasion assay,cell apoptosis test,were used to observe the changes of proliferation,migration,invasion,apoptosis and other biological characteristics of A549 cells and JT cells,so as to explore the mechanism of miR-551b-3p regulating the growth of lung cancer.Results: The expression of miR-551b-3p in lung cancer patients of Xuanwei area,Yunnan Province was significantly lower than that in the corresponding para-carcinoma lung tissues(P<0.001).The expression level of miR-551b-3p was negatively correlated with tumor diameter(P=0.005<0.05).Among 52 patients,there were 37 patients whose miR-551b-3p expression level in preoperative serum samples was lower than that in postoperative serum,and 15 patients whose miR-551b-3p expression level in preoperative serum samples was higher than that in postoperative serum,however,the expression level of miR-551b-3p in preoperative serum samples has no significant difference with that in postoperative serum for the total 52patients(P=0.308>0.05).Compared with BEAS-2B,the expression level of miR-551b-3p in A549 cells was decreased,the difference was statistically significant(P<0.01).The expression level of miR-551b-3p in JT cells was decreased,and the difference was statistically significant(P<0.05).Compared with the Control group and the NC group,A549 cells and JT cells in the miR-551b-3p mimics group were inhibited in cell proliferation assay,plate clone formation experiment,cell scratch test,Transwell chamber migration and invasion assay,while in the cell apoptosis test,the number of early apoptotic cells was significantly increased.The difference was statistically significant(P <0.001).Conclusion: The expression of miR-551b-3p is low in lung cancer tissues of Xuanwei area,Yunnan Province,the larger the tumor diameter,the lower of the expression level of miR-551b-3p,miR-551b-3p may be a tumor-inhibiting miRNA in lung cancer of Xuanwei area,Yunnan Province,whereas the expression of miR-551b-3p in serum samples has no statistical difference.In the cell biological function experiment,miR-551b-3p over-expression could inhibit the proliferation,the invasion and migration ability of A549 cells and JT cells,and promote the apoptosis of A549 cells and JT cells,miR-551b-3p plays a role of tumor-inhibiting miRNA.So miR-551b-3p can possibly inhibit the development of lung cancer in Xuanwei area,Yunnan Province,it provides a certain theoretical basis and direction for the mechanism further study of lung cancer in Xuanwei area. |
