Objective:To establish an animal model of subcutaneous transplanted tumor and tail vein injection lung metastasis in nude mice with NEAT1 overexpression and normal expression of human hepatocellular carcinoma cells,and to detect the effect of NEAT1 overexpression on PIM1,MYC,ATF4 and MAPK1 expression in the subcutaneous tumor model in nude mice,and then further investigate the effect of NEAT1 overexpression on the growth and metastasis of human HCC cells in nude mice,and to provide a new target and idea for The study will provide new targets and ideas for the diagnosis and treatment of hepatocellular carcinoma.Methods : 1.Lentiviral transfection of human hepatoma cell SK-HEP-1,puromycin screening to establish NEAT1 overexpression and normal expression stable cell line.2.Establish subcutaneous xenograft models of nude mouse liver cancer with normal and overexpression of SK-HEP-1 cells of NEAT1 and lung metastasis model by tail vein injection,and regularly weigh and measure the size of subcutaneous transplanted tumors during the experiment.Regularly observe the activity status,body weight and diet of nude mice.3.The effect of Lnc RNA NEAT1 on the growth and metastasis of human hepatoma cells in nude mice was explored by constructing a subcutaneous xenograft model and injecting lung metastasis model through the tail vein of nude rats,and q RT-PCR and Western Blotting detected the expression of PIM1,MYC,ATF4 and MAPK1 in subcutaneous transplant tumor tissues.Results:1.Successfully established a subcutaneous xenograft model for nude mouse liver cancer.2.A lung transfer model was successfully established through nude mouse tail vein injection.3.HCC cells overexpressing NEAT1 promoted the growth of subcutaneous xenograft tumors in nude mice.4.HCC cells overexpressing NEAT1 promoted lung metastasis in nude mice.5.Upregulation of PIM1,MYC,ATF4,MAPK1 gene expression in subcutaneous xenograft tumors of nude mice overexpressing NEAT1 in HCC cells.Conclusion:Overexpression of NEAT1 promoted the growth and metastasis of human hepatocellular carcinoma cells in nude mice,and upregulated the expression of PIM1,MYC,ATF4,and MAPK1 genes.
