Objective:To explore the pathologic characteristics and mechanism of antibody-mediated rejection(AMR)in sensitized rats after reduced liver transplantation.Methods:Recipient:Lewis rats,250-290g,10-12 weeks of age;Donor:Brown Norway rats,250-280g,10-12 weeks of age.Spleen cells were obtained from BN rats by grinding,cell counter counts 5×10~7cells and then they were injected from caudal vein of Lewis rats,after two weeks,Lewis rats accept volume reduction liver transplantation.The left outer lobe of the BN rats liver was isolated and clipped,this part of the liver accounting for about 32%-35%of the total volume of the liver.According to“the two-sleeve method”,establish the reduced volume rat liver transplantation model.Blood and fresh liver tissue were collected after 14 days of transplantation,serum samples were obtained by blood centrifugation and analyze the aspartate aminotransferase(AST),alanine aminotransferase(ALT),total bilirubin(TB)and creatinine(CRE).The expression of CD3,CD68 and CD86 were detected by immunofluorescence.Hematoxylin-eosin(HE)and Masson staining were used to evaluate the liver histopathological indexes;CK-19,C4d and CD20 used immunohistochemical staining methods to detected.T test or ANOVA were perfoemed for data between different groups according the characteristics.Result:(1)Rejection occured in the whole liver transplantation group of rats after liver transplantation,and the characteristic of rejection were consistent with the pathological diagnostic criteria of AMR;(2)Compared with the whole liver transplantation group,the reduced volume liver transplantation group showed more obvious inflammatory cell infiltration,vascular endothelial injury,microvascular inflammation,biliary tract destruction and other antibody mediated rejection reactions.In immunofluorescence,CD3 and CD68 were mere strongly expressed.Conclusion:After reduced volume liver transplantation in sensitized rats,liver rejection was more severe acute rejection and transplantation liver dysfunction occurred.and the acute rejection in sensitized rats was consistent with the characteristics of AMR.
