| Objective: By detecting the expression levels of Lnc RNA H19 and mi R-129 genes in peripheral blood of patients with Amnestic mild cognitive impairment(a MCI),the relationship between Lnc RNA H19 and mi R-129 and a MCI was explored.To provide theoretical basis for further screening biological markers and therapeutic drug targets in early diagnosis of a MCI patients.Methods: 1.According to the inclusion criteria,78 elderly patients aged 60-85 years old who visited the Department of Neurology and/or the memory and Parkinson clinic,physical examination center and community Hospital of the First Affiliated Hospital of Jiamusi University from December 2021 to December 2022 were selected.Among them,38 patients(14 males and 24 females)were enrolled in the a MCI group.The mean age was(68.87±5.51)years,and 40 patients(19 males,21 females,mean age 70.40±4.96 years)were enrolled in the NC group.2.Complete neuropsychological evaluation,Clinic Dementia Rating(CDR),Hamilton depression scale(HAMD),Hamilton Anxiey scale(HAMA),Hachinski ischemic scale(HIS),Mini Mental State Examination(MMSE),Montreal Cognitive assessment(Mo CA),Clockdrawing Test(CDT),Boston naming test(BNT),Auditory verball earingtest(AVLT),Trail making test(TMT),and Activity of daily living(ADL)were used to assess cognitive function and mental behavior.3.5ml of fasting venous blood was collected from all subjects in the morning of the next day after inclusion,and the expression levels of Lnc RNA H19 and mi R-129 genes in peripheral blood of the two groups were measured by RT-q PCR.4.The expression levels of Lnc RNA H19 and mi R-129 genes in the two groups were respectively compared.Receiver operating characteristic curve(ROC)was used to analyze the diagnostic value of Lnc RNA H19 and mi R-129 for a MCI.Pearson correlation analysis was used to discuss the correlation between Lnc RNA H19 and mi R-129 in the a MCI group,and SPSS29.0 software was used for data processing.Results: 1.Comparison of Lnc RNA H19 expression level in peripheral blood between the two groups: Lnc RNA H19 expression level in a MCI group was higher than that in NC group,the difference was statistically significant(P < 0.05).2.Comparison of the expression level of mi R-129 in peripheral blood between the two groups: the expression level of mi R-129 in a MCI group was lower than that in NC group,and the difference was statistically significant(P < 0.05).3.The scores of MMSE,MOCA,CDT,AVLT delayed recall and BNT in a MCI group were lower than those in NC group,with statistical significance(P < 0.05),while the scores of TMT-A and TMT-B in a MCI group were higher than those in NC group,with statistical significance(P < 0.05).4.Pearson correlation analysis showed that Lnc RNA H19 and mi R-129 gene expression levels in serum of a MCI group were negatively correlated(r=-0.417,P < 0.05).5.The area under curve(AUC)of Lnc RNA H19 for the diagnosis of a MCI was 0.750,95% confidence zone(0.639-0.841),sensitivity was 73.68%,specificity was 72.50%.The area under the curve(AUC)of mi R-129 for the diagnosis of a MCI was 0.774,the 95% confidence interval was(0.666-0.861),the sensitivity was 76.32%,and the specificity was 67.50%.Conclusions:1.Lnc RNA H19 and mi R-129 are related to the occurrence and development of a MCI diseases,and their expressions in a MCI diseases are negatively correlated.2.Both Lnc RNA H19 and mi R-129 have certain sensitivity and specificity for the diagnosis of a MCI,but whether they can be biological markers of a MCI remains to be further studied. |
