| ObjectiveThe research is conducted from renal fibrosis and renal damage which is caused by renal tubular epithelial cell apoptosis induced by hypertensive renal damage of renal ischemia and hypoxia.Through the observation of Jiangya Tongluo Formula on rat renal tubular epithelial cells(NRK-52E)apoptosis effect induced by ischemic hypoxia,this paper attempts to explore the mechanism of hypertension renal damage by way of Jiangya Tongluo Formula,providing a molecular biological foundation for the clinical application of Jiangya Tongluo Formula to hypertension renal damage.Method42 male SD rats(weight 200+20g)were randomly divided into 3 groups:control group 18,Valsartan group 10,Jiangya Tongluo Formula group 14.These rats were given normal saline,Valsartan,Jiangya Tongluo Formula by gavage respectively for 7 days before intraperitoneal anesthesia,drawn blood from abdominal aortic to prepare containing serum in vitro cultivation.NRK-52E were cultured in vitro to the normal growth before ischemia hypoxia for 6 hours,8 hours,10 hours,12 hours.The cell viability was measured by Cell Counting Kit-8(CCK8)to calculate the apoptotic rate.On the condition of ischemia and hypoxia for 12 hours,apoptosis of the NRK-52E model was established.The cells were divided into control group,model group,Valsartan group,Jiangya Tongluo Formula group.The last two groups were given Valsartan and Jiangya Tongluo Formula containing serum medium intervention;while the first two groups were given normal rat serum medium.The cell viability of the above groups was measured by CCK-8 after 10 hours,14 hours,and 18 hours respectively,on the basis of which the cell relative apoptosis rate was calculated.Results1.Ischemia and hypoxia for 6 hours,8 hours,10 hours,and 12 hours will lead to NRK-52E apoptosis.There was a statistically significant difference compared with the control group(P<0.05,P<0.01).2.After drug intervention,the cell viability has increased and apoptosis rate has decreased.The result was statistically significant(P<0.01)compared with the model group.As time passed by and the effect strengthened,there was a statistically significant difference with an intervention for 10 hours(p<0.01)instead of 14 hours and 18 hours(p>0.05).Jiangya Tongluo Formula gave preferential treatment than valsartan group,but has no significant difference.The improvement of cell apoptosis rate would be at the top in 14 hours and such condition would not change remarkably even if time was extended.Conclusion1.Ischemia and hypoxia can induce the apoptosis of NRK-52E,and is related with time.2.Jiangya Tongluo Formula can significantly inhibit the apoptosis of NRK-52E induced by ischemia and hypoxia.3.Jiangya Tongluo Formula can inhibit the apoptosis of renal tubular epithelial cells of hypertensive renal damage due to hypoperfusion,reduce kidney damage and protect renal function to delay the process of renal damage of hypertension. |
