"Formula-pattern elements correspondence" is a principle for prescribing formula.It is considered that prescribing formula according to the correspondence between "formula element" and "pattern element",which belongs to one kind of thinking model of prescribing clinical formula and emphasizes "principle-method-recipe-medicines" logical relationship more.It is a principle which should be followed during changing formula.It can make"formula element" and"pattern element" one-to-one correspondence,the targeting of the medicine in the formula clearer and treatment more individual and accurate.Non-alcoholic fatty liver disease is a clinical pathological syndrome,which characteristics is steatosis of hepatocytes and fat storage in the liver.The case rate of it increases every year in China and other countries in Asia and the West.It also becomes the most common cause of chronic liver disease all over the world.ObjectiveThe research involved in three parts:review,theoretical discussion and experiment research.We copied NAFLD rats model and observed the variety of every group rats fecal water content movements and intestinal flora,aiming to observe the effects in "eliminating the turbid by purgation" formula element in Da Chaihu decoction on eliminating the turbid by purgation.As a result,it can provide experiment evidence for understanding the effect of theueliminating the turbid by purgation" formula element in Da Chaihu decoction and recognizing the structure of Da Chaihu decoction and its mechanism of treating NAFLD.MethodsWe used the method of high fat and high sugar diet inducing 16 weeks continuously to copy and evaluate the non-alcoholic fatty liver disease rat model.After that,we use separately "eliminating the turbid by purgation"formula element in Da Chaihu decoction and Da Chaihu decoction to treat NAFLD rats.We observed the effects in "eliminating the turbid by purgation" formula element in Da Chaihu decoction on eliminating the turbid by purgation from the point of macroscopic view by dynamic observing the variety of every group rats fecal water content movements.We observed the effects in"eliminating the turbid by purgation" formula element in Da Chaihu decoction on eliminating the turbid by purgation from the point of microcosmic view by dynamic observing the variety of every group rats intestinal flora.Result1.Effect on fecal water content in NAFLD rats①General condition:In the normal group,the rats grew well with coat luster and good mental condition.There were no abnormality in the defecation.Compared with the normal group,the rats in the model group had lack of luster,rough scales,and irritability.Their fecal volume was reduced with dry fecal pellets,shortened pellet shape,and difficulty in drainage;Compared with the model group,rats in the "eliminating the turbid by purgation" formula element group and Da Chaihu decoction group had increased hair luster,reduced irritability symptoms,increased fecal volume,softened fecal pellets,granular growth and easy to discharge.②Model evaluation:Compared with the normal group,the liver index of the model group rats was significantly higher(P<0.01);HE showed that in the model group,the central vein and the middle of the portal area were stained shallowly,the hepatocyte cords were arranged disorderly,the hepatocytes were swollen,some hepatocytes showed a ballooning change,and lipid vacuoles of various sizes were seen in the cytoplasm.The nucleus is squeezed to one side;Compared with the normal group,the ALT and AST levels in the model group were significantly higher(P<0.01).③Fecal water content:During the administration,the fecal water content in the model group rats was significantly lower than that in the normal group(P<0.01).In the first 4 days of administration,compared with the model group,there was no significant change in the fecal water content of the rats in the administration group,but there was a tendency of increase in the "eliminating the turbid by purgation" formula element group,the Da Chaihu decoction group first decreased and then increased.On the 6th to 8th days of administration,compared with the model group,the fecal water content of the rats in the the "eliminating the turbid by purgation" formula element group was significantly higher(P<0.05).The Da Chaihu decoction group had an increasing trend,but there was no difference;and compared with the Da Chaihu group,there was a trend of increasing the "eliminating the turbid by purgation" formula element group.On the 10th to 14th days of administration,the fecal water content in the two groups was increased compared with the model group,and was significantly higher on the 12th day than the model group(P<0.01);Basically the same,the "eliminating the turbid by purgation"formula element group was slightly higher.On the 16th to 18th day of administration,the fecal water content of the rats in the"eliminating the turbid by purgation" formula element group was significantly higher than that in the model group(P<0.05),and peaked on the 18th day;the Da Chaihu decoction group has a trend of increasing,but a downward trend compared to before.Compared with the Da Chaihu decoction group,there was a tendency for the "eliminating the turbid by purgation" formula element group to increase.On the 20th to the 22nd day of administration,compared with the model group,the fecal water content of the rats in the two groups had a tendency to increase,but it was in a decreasing state compared with the previous period;compared with the Da Chaihu decoction group,there was a trend of increasing the "eliminating the turbid by purgation" formula element group.On the 24th to 28th days of administration,the fecal water content of the rats in the two groups was significantly higher than that in the model group(P<0.05),and was higher than before.Compared with the Da Chaihu decoction group,there was a tendency for the"eliminating the turbid by purgation" formula element group to increase.2.Effect on intestinal microflora in NAFLD rats:① At the phylum level,there was no significant difference in the relative abundance of Firmicutes and Bacteroidetes in the intestinal flora between the model group and the normal group.Compared with the model group,the relative abundance of Firmicutes in the intestinal flora of the rats in the "eliminating the turbid by purgation" formula element group was decreased(P<0.05),and the relative abundance of Bacteroidetes had a trend of increasing;There was no significant difference in the relative abundance of Firmicutes and Bacteroidetes in the intestinal flora of the Da Chaihu decoction group.Compared with the Da Chaihu decoction group,the relative abundance of Firmicutes in the intestinal flora of the"eliminating the turbid by purgation" formula element group decreased,and the relative abundance of Bacteroidetes increased.②At the genus level,compared with the normal group,the relative abundance of Bacteroidetes and Blautia of the model group increased(P<0.05),and the relative abundance of Lactobacillus decreased(P<0.01).Compared with the model group,the relative abundance of Bacteroidetes and Sutterella of the "eliminating the turbid by purgation" formula element group was increased(P<0.05).Compared with the model group,the relative abundance of Sutterella of the Da Chaihu decoction group was increased(P<0.05).Compared with the Da Chaihu decoction group,the relative abundance of Bacteroidetes of the"eliminating the turbid by purgation" formula element group was increased(P<0.05),and Sutterella has an increasing trend.③ Specific group of bacteria:The specific group of bacteria in the normal group are Ruminococcaceae,Ruminococcus,Oscillospira,Lactobacillales,Lactobacillaceae,Lactobacillus,Bacilli,TM7,Spirochaetes,Clostridium,Elusimicrobia,Elusimicrobiales,Lactoba cillu shelveticus,Lactobacillu sreuteri,Elusimicrobium,RF39,Christensenellaceae.The specific group of bacteria in the model group was Ruminococcusgnavus.The specific bacterial flora of the "eliminating the turbid by purgation" formula element group includes Bacteroidaceae,Bacteroides,Porphyromonadaceae,Parabacteroidea,4C0d2,Cyanobacteria,YS2,Parabacteroidesdistasonis,Bacteroidesuniformis,Verrucomicrobiae,Verrucomicrobiales,Verrucomicrobiaceae,Akkermansia,Akkermansia muciniphila,Coprobacillus,Parabacteroides-gordonii,Blautia-producta,Ruminococcustorques.The specific group of bacteria in the Da Chaihu decoction were Erysipelotrichi,Erysipelotrichales,Erysipelothrichaceae.ConclusionsThe research finds that"eliminating the turbid by purgation" formula element in Da Chaihu decoction can improve dry stool and intestinal flora in the NAFLD rats.It is believed that the target of"eliminating the turbid by purgation" formula element in Da Chaihu decoction treating NAFLD is the intestinal.In addition,it has close contacts with intestinal flora.The research also provides experiment evidence for recognizing further the formula structure of Da Chaihu decoction and clinical application.
