| PurposeThe purpose of this study was to determine TCM syndromes in rats with NAFLD induced by a high-sugar high-fat diet.This research paper is elaborated in three parts:literature review,theoretical discussion and experimental research.The literature review summarizes the advances in the pathogenesis,diagnosis and treatment of Chinese and Western medicine for non-alcoholic fatty liver disease,and the research progress in animal model collection and syndrome determination in Chinese medicine.It provides relevant theoretical support for animal experiments.The theoretical discussion is mainly based on the basic theory of traditional Chinese medicine to explore the formation of non-alcoholic fatty liver disease and the mechanism of non-alcoholic fatty liver disease.The contents of the experimental study included the construction of a rat model of NAFLD induced by a high-sugar high-fat diet and the determination of the basic syndromes of Chinese medicine in animal models.MethodFirst,the NAFLD rat model was constructed using a sustained high-glucose and high-fat diet for 12 weeks.Second,in the process of constructing the model,the animal’s macroscopic signs were dynamically acquired.The macroscopic signs obtained were correlated with the clinical NAFLD syndromes.The homogeneity of the models was compared,and the basic syndrome attributes of the model animals were initially determined.Third,based on the obtained basic syndrome attributes,the liver irritability scores,the phlegm moisture scores,and the hot junction scores were respectively established.Finally,according to the selected Dachaihu decoction,the relevant performances of the model group and the drug-administered group were compared after four weeks of treatment,and the effect of Dachaihu Decoction on the pathological model of NAFLD was observed.Results(1)Model construction:HE staining showed that hepatocytes in the hepatic tissue of the model group were swollen,part of the hepatocytes were ballooning,the nucleus was squeezed to the side,and the size of the liver cells could be seen in most liver cells.The cables are in disorder.Oil red O-stained frozen sections showed that the oil red O of the model group was markedly colored,covering almost all hepatocytes under the microscope.AST and ALT measurements showed that the AST and ALT values of the model group were significantly higher than the normal group,indicating that the liver function of the model group had a certain degree of change compared with the normal group.The liver index showed that the liver index of the model group was significantly higher than that of the normal group.(2)TCM syndrome identification:The macroscopic signs of the animal model showed that the rats in the normal group had a good overall condition during the 16-week period,and no obvious abnormalities were observed;the rats in the model group appeared physical wasting,mental irritability,and frightened each other during 6 weeks to 16 weeks.,Hyperactivity,red eyes,hair yellow,hard stool,urine loss and other abnormal performance.The syndrome scores showed that all rats in the model group had a score of liver qi of>1.5;in the normal group,only one rat had a liver qi score of>1.5,and the rest were<1.5,indicating that the model group did have liver qi stagnation;normal rats The phlegm scores were mostly maintained at about 1 minute,and the fluctuations did not exceed 0.5,and their scores were all<1.5;in the model group,there were three rats with a phlegm score>1.5,which could be judged as dampness syndromes.The others were Between 1 and 1.5,no judgment can be made;because the number of dampness syndromes in the model group is only a few,it is considered that the performance of the dampness syndrome in the model group is not obvious;The score remained at about 1 point,up and down no more than 0.5,and their scores were all<1.5,while the scores in the model group were significantly>1.5,all were between 1.8 and 1.9.The hot knot score was high,so the model group was judged.Heat syndrome is present in rats.The results of the Israeli test showed that in the general form,the rats of the model group were emaciated,had violent reaction,abnormal irritability,frequent movements,frequent fighting and fighting,severe resistance during grabbing,red eyes,yellowish coat,and dietary water intake.Reduced,hard and dry bowel movements,yellow urine;while the rats in the administration group were lean,responsive,irritated and relieved,eyes pale red,yellow hair,diet water increased to a certain extent,stool is hard,urine Small amount;on the pathological picture,HE stained sections showed that the liver tissue in the administration group was significantly improved compared with the model group,and the fat drops were significantly reduced compared with the model group,the liver sinus was clearer,and the hepatic cord was arranged more neatly.Oil red O staining showed that the oil red O staining of the administration group was reduced compared with the model group,and the red area coverage was also reduced compared with the model group.Conclusions(1)Establishment of NAFLD rat model:A rat model of NAFLD can be established successfully by simply feeding on a high-sugar and high-fat diet.Observed in animal models,it was found that the simple taste of fat and sweet can also injure the liver and lead to the occurrence of NAFLD.(2)The TCM Syndromes of Rats with High-sugar and High-fat-induced NAFLD Model:Through the observation of macroscopic characterization of the model rats,clinical homogeneity and syndrome scores,the TCM syndromes of rats with NAFLD induced by high-sugar high-fat diet were determined as Liver enthusiasm enthusiasm;and through a theoretical study to explain the simple diet caused by liver enthusiasm hot junction type NAFLD TCM pathological mechanism:eating fat and thick greasy liver,damage the spleen and stomach,loss of transport of the Secretary,dampness endogenous;In the middle coke,which leads to blockage of the gas engine,blood flow is not smooth,which in turn leads to dampness and stasis;dampness,stasis,and stasis,will further lead to liver qi stagnation,liver and stomach sensation of heat,resulting in Yangming fever knot,muddy Thermal injury to the liver,liver and intestines formed a vicious circle,leading to the occurrence of liver jaunder-heat type NAFLD.(3)Observe the effect of Dachaihu Decoction on the pathological model of NAFLD:Dachaihu Decoction can improve the performance of macroscopic signs such as irritability,hyperactivity,and body wasting in model animals;in pathological morphology,HE staining of model animals after Dachukang decoction administration The liver tissue was significantly improved.The hepatic cord was arranged more neatly than the model group.Fat droplets were significantly reduced compared with the model group.Oil red O staining showed that the oil red O area in the administration group was significantly reduced compared with the model group.The red color area coverage is also significantly reduced compared to the model group. |
