Objective:In vivo experiments were conducted to study the effects of Sanjie Xiaonao prescription on transplanted tumors in mice with breast cancer,aiming to explore the efficacy and mechanism of Sanjie Xiaonao prescription.Methods: 1 、 Mice transplanted tumor models of breast cancer mice were established.All the transplanted tumor models of breast cancer mice were randomly divided into five experimental groups: model group,low-dose group of Sanjie Xiaonao prescription,medium-dose group of Sanjie Xiaonao prescription,high-dose group of Sanjie Xiaonao prescription,and positive control group,with 9 mice in each group.2、 The mice were given drugs continuously for 24 days.The diet and activity of the mice were observed.The changes of body weight and tumor volume of the transplanted mice were recorded and the tumor inhibition rate was calculated.3、 He staining was used to detect the morphology of the transplanted tumor tissue cells in each group,and TUNEL staining was used to detect the apoptosis of the transplanted tumor tissue cells in each group.4、 Western blot was used to detect the expression of epithelial-related proteins(E-cadherin,N-cadherin,Vimentin)and the content of transcription factors Twist and Snail in the transplanted tumor tissues of mice in each group.Results: 1、The food intake and activity status of mice treated with Sanjie Xiaonao prescription were less affected by the transplanted tumor of breast cancer.2 、 After calculation,compared with the model group,there were statistically significant differences in the volume and weight of the transplanted tumor body in the administration group(P < 0.01),and the volume of the transplanted tumor body in the medium dose group of Sanjie Xiaonao Formula was smaller than that in the low dose group,with statistically significant differences between the groups(P < 0.05).The tumor inhibition rates among different administration groups were 52% in the medium dose group of Sanjie Xiaonao prescription,40% in the positive control group,39% in the low dose group of Sanjie Xiaonao prescription,and 34% in the high dose group of Sanjie Xiaonao prescription.3、Most of the transplanted tumor bodies in each group showed round,quasi-round or lobulated growth.Under light microscope,the tumor cells were slightly enlarged,the atypia cells were reduced and the tumor tissue structure was destroyed in the treatment group.TUNEL staining results showed that compared with model group,the apoptosis index of drug administration group was significantly different(P < 0.05).4、 Western blot results showed that compared with model group,the expression levels of epithelial-related protein E-cadherin in transplanted tumor tissues of mice in drug administration group were increased,the expression levels of N-cadherin and Vimentin were decreased,and the levels of transcription factors Twist and Snailin in tissues were decreased,with statistical significance(P < 0.05).Conclusion: 1、Sanjie Xiaonao prescription can improve the weakness and reduced food intake of mice with breast cancer tumor transplantation.2 、 Sanjie Xiaonao prescription can inhibit the growth of tumor volume to a certain extent,and the inhibition rate of medium dose of Sanjie Xiaonao prescription is better than that of low and high dose Sanjie Xiaonao prescription groups.3 、 Sanjie tumor-elimination prescription can increase the apoptosis rate of tumor tissue in mice transplanted with breast cancer,and the effect of Sanjie tumor-elimination prescription in medium dose is more obvious,it can promote the apoptosis of tumor cells.4 、 Sangjiejiaoxiao prescription can inhibit the expression of transcription factors Twist and Snail in the transplanted tumor tissues of breast cancer mice,and up-regulate the expression level of E-cadherin protein in the transplanted tumor tissues of breast cancer mice,while down-regulate the expression levels of N-cadherin and Vimentin protein.Sanjie Xiaonao Formula can reduce the occurrence of epithelial mesenchymal metastasis of breast cancer,which is considered to be related to epithelial mesenchymal transformation.
