Objective:This study observed the effects of"xingnaokaiqiao" acupuncture method,Buy ang Huanwu Decoction,"xingnaokaiqiao" acupuncture method combined with Buy ang Huanwu Decoction on the area ratio of cerebral infarction and nerve cell apoptosis related factors in brain tissue of MCAO model rats,and conducted network target topology analysis to explore the advantages of combined acupuncture and medicine compared with simple acupuncture and traditional Chinese medicine and the possible mechanism of treating cerebral infarction.Methods:The experimental groups were divided randomly.250 SPF grade s-day rats(half male and half female)were randomly divided into sham operation group(50 rats)and model group(200 rats).The model group was prepared according to the middle cerebral artery embolization(MCAO).After the model was established,the rats were scored in behavior and the successful animals were randomly divided into model group(50 rats),acupuncture group(5 0 rats),traditional Chinese medicine group(5 0 rats)and combined acupuncture and medicine group(5 0 rats).The sex of the five groups was half male and half female.The sham operation group and acupuncture drug group were treated with acupuncture and traditional Chinese medicine by gavage,the acupuncture group was treated with acupuncture,the traditional Chinese medicine group was treated with traditional Chinese medicine by gavage,and the model group was not treated.The acupuncture operation was 20min each time,twice a day.The traditional Chinese medicine group was only bound without acupuncture.Traditional Chinese medicine was administered by gavage once a day at the rate of 0.6ml/100g.The above four groups were divided into five time points:1 day,3 days,5 days,7 days and 14 days.10 rats were randomly selected from each group at each time point.4 rats were randomly selected to detect the infarct area of brain tissue,and 6 rats were randomly selected to detect the effects of brain cell necrosis and apoptosis related factors(AIF,Bcl-2,Bax,Caspase-3,chop,Fas,FasL,p-JNK,p53)only by ELISA.The contents of 9 targets detected by ELISA kit were calculated and the mutual network relationship between targets was constructed.Result:1.The area ratio of cerebral infarction:According to the overall data,the area ratio of cerebral infarction in the acupuncture and drug group was smaller than that in the model group,acupuncture group and traditional Chinese medicine group,and the difference was statistically significant(p<0.05).The area ratio of cerebral infarction decreased gradually with time,and the area ratio of cerebral infarction was the smallest at 14 days after operation.On the 1st,3rd,5th and 7th day after operation,there was no significant difference between the acupuncture group and the traditional Chinese medicine group at the same time point(p>0.05).At 14 days after operation,the difference between the acupuncture and drug group,the acupuncture group,the traditional Chinese medicine group and the model group was statistically significant(p<0.05).2.Detection of apoptosis related indicators:①Within 14 days after operation,the acupuncture drug group can significantly reduce the content of apoptosis related factor Bax compared with the traditional Chinese medicine group.There is no significant difference in the content of AIF,Bcl-2,Caspase-3,chop,Fas,FasL,p-JNK and p53 in the brain tissue of the acupuncture drug group compared with the acupuncture group and the traditional Chinese medicine group.②The indexes of AIF,B ax,Caspase-3,Fas,FasL,p-JNK and p5 3 showed a downward trend with time after operation.Among them,the contents of Fas and p53 in the brain tissue of the acupuncture and drug group were significantly different among the groups(p<0.05),and the changes of Fas and p53 showed a significant downward trend with time.The contents of AIF,Bax,Caspase-3,FasL and p-JNK in brain tissue of acupuncture and drug group had no significant difference with time(P>0.05).3.Topology analysis of network targetBy calculating the contents of 9 targets detected by ELISA kit and constructing the mutual network relationship between targets,it was found that Bcl-2 had the strongest effect,followed by Fas,and caspase-3 had the smallest effect.Fas has the largest action coefficient,and the action coefficients of other targets are relatively discrete.The cumulative percentages of AIF,Bax,Bcl-2,chop,Fas,FasL,p-JNK and p53 are roughly the same,while the cumulative percentages of Caspase-3 are relatively low.AIF,Bax and Fas were negative,and the other six targets were positive.Among the network relationships among targets,baxfasl has the strongest positive relationship and chop-p53 has the strongest negative relationship.Conclusion:1."Xingnao Kaiqiao" acupuncture combined with Buy ang Huanwu decoction may reduce the area ratio of cerebral infarction by inhibiting cell apoptosis,and its curative effect is better than that of "Xingnao Kaiqiao" acupuncture alone and Buyang Huanwu Decoction;2.Xingnao Kaiqiao acupuncture combined with Buyang Huanwu decoction may inhibit apoptosis after cerebral infarction by down regulating AIF,bax/bcl-2,Caspase-3,chop,Fas,FasL,p-JNK and p53,among which Bcl-2,Fas and Bax may be the key targets for the combination of acupuncture and medicine to inhibit apoptosis.3.Combined with the pathological mechanism of cerebral infarction,it was found that each target had different key time periods of interventional acupuncture and traditional Chinese medicine treatment.4.Combined with the study of network topology,the parameter correlation coefficients and eigenvalues between targets reflect their different positions and roles in the target network.The nine targets have their own independent roles and are interrelated.The above provides some ideas for the future research on the relevant mechanisms of acupuncture and medicine in the treatment of cerebral infarction,and provides a certain basis for the modernization of acupuncture research and the development of new drugs.
