Purpose: Corneal ulcers are a blinding disease with a high incidence and difficult treatment in the field of ophthalmology.In the experiments of this thesis,corneal ulcers were treated by Chinese and Western medicine therapy of autologous blood spotting and calf blood deprotein extract ophthalmic gel drops,aiming to compare the repair results of corneal ulcers by different methods of treating corneal ulcers,from a macroscopic perspective,corneal inflammation as well as trauma scoring;from a microscopic perspective,the levels of MMP-1 and TIMP-1 were measured simultaneously,and the two perspectives were cut separately,which can make the experimental results more accurate and authoritative,which can provide a simple and reliable treatment plan for clinical ophthalmology,and find an effective method to treat corneal ulcers.Materials and methods: In this experiment,36 Japanese Large-eared White rabbits were randomly divided into 4 groups after the establishment of a corneal ulcer model by using a corneal ring drill.Each group of 9 experimental rabbits was divided into group A blank control group(no corneal excision),group B experimental group,group C positive control group and group D negative control group.The extent of corneal ulcer repair was observed on days 3,7 and 14 during the 14-day experimental period.The samples were removed for testing on the last day and the data of MMP-1 and TIMP-1 were measured using the WB assay.Results:1.Compared to groups A and D,both groups B and C showed a significant reduction in the area of corneal ulcer trauma after 14 days,and the treatment had a significant effect.2.The MMP-1 content in group B was much lower than that in group D.The difference was statistically significant,with P < 0.01.The MMP-1 content in group B was also lower than that in group C,with P < 0.01,which was statistically significant.3.At the same time,the TIMP-1 values were consistently higher in groups B and C compared with groups A and D.The difference was statistically significant,P <0.01;the TIMP-1 content in group B was more than that in group C,P < 0.01,which was statistically significant.Conclusion:1.Both autologous blood therapy and ophthalmic gel of calf blood deproteinized extract can treat corneal ulcers.2.Compared with calf blood deprotein extract ophthalmic gel,autologous blood therapy was more effective in reducing the level of MMP-1 in the cornea,inhibiting the corneal inflammatory response and speeding up the recovery of the cornea.3.Compared with calf blood deproteinized extract ophthalmic gel drops,autologous blood therapy increased the level of TIMP-1 in the cornea and achieved the efficacy of promoting the stability of the outer collagen fibres of corneal stromal cells,which could inhibit the continued development of corneal ulcers.
