| Objective: To investigate the effect of Yiqi Yangyin Huoxue Tongluo Recipe on cardiac function and the expression of AMPK and ULK1 proteins in rats with acute myocardial infarction,and to explore the potential drug targets of integrated traditional Chinese and Western medicine in the treatment of cardiovascular diseases.Materials and methods: 60 SD male rats were randomly divided into 6 groups with 10 rats in each group.They were divided into blank control group,model group,high-dose Shuangshen Huoxue Granules group,Shuangshen Huoxue Granules medium dose group,and Shuangshen Huoxue Granules group.Huoxue Granules low-dose group and trimetazidine hydrochloride group.After the rats were preconditioned for 1 week,modeling was started.The rat model of acute myocardial infarction was prepared by ligating the left anterior descending coronary artery in the experiment.The blank control group and the model group were given 3 ml of normal saline daily,and the dosage of the drug group was converted by referring to the body surface area ratio of commonly used experimental animals and humans.The middle dose group of granules was 3.96g/kg,the low dose group of Shuangshen Huoxue Granules was 1.98g/kg,and the trimetazidine hydrochloride group was 1.90mg/kg,and each of the above groups was intragastrically administered once a day.After continuous intragastric administration for 2 weeks,the rats were detected by cardiac color Doppler ultrasonography,and LVIDs,LVIDd,LVPWd,LVPWs,LVEF and LVFS were recorded;the rat hearts were harvested,and the protein expression levels of AMPK and ULK1 in the rat myocardium were detected by WB method.Results:1 General status observationThe observation results showed that,compared with the blank control group,the rats in the model group had rough,dull,sparse hair,slack spirit,slow movement and loss of appetite.Compared with the model control group,the hair of the rats in the medication group regained luster,the movements were flexible,and the state of mind and appetite was good.2 rats died in the model group,and 1 rat died in the Shuangshen Huoxue Granules middle-dose group.2 Comparison of the results of cardiac color Doppler ultrasound in each group of rats2.1 Left ventricular end-systolic diameters(LVIDs)and left ventricular end-diastolic diameters(LVIDd)in ratsThe results showed that compared with the blank control group,the LVIDs and LVIDd of the rats in the model group were significantly increased(P<0.01).Compared with the model group,the LVIDs and LVIDd of the rats in the Shuangshen Huoxue Granules(high,medium and low doses)group and trimetazidine hydrochloride group decreased(P<0.05).There was no significant difference in LVIDs and LVIDd between the high,medium and low dose groups of Shuangshen Huoxue Granules(P>0.05).There was no significant difference between the drug groups.2.2 Comparative analysis of the thickness of the posterior wall of the left ventricle in each group of ratsThe results showed that compared with the blank control group,LVPWd and LVPWs in the model group were significantly increased(P<0.01).Compared with the model group,the LVPWs of the rats in the high-dose Shuangshen Huoxue Granules group and the trimetazidine hydrochloride group decreased(P<0.05).There was no significant difference between the drug groups.2.3 Comparison of the results of left ventricular ejection fraction(LVEF)and left ventricular short-axis rate(LVFS)in ratsThe results showed that compared with the blank control group,the LVEF and LVFS of the model group were significantly decreased(P<0.01).Compared with the model group,the LVEF and LVFS of the rats in the Shuangshen Huoxue Granules(high,medium and low doses)group and trimetazidine hydrochloride group were increased(P<0.05).There was no significant difference between the drug groups.3 Western Blotting method to detect the protein expression levels of AMPK and ULK1The results showed that compared with the blank control group,the expression levels of P-AMPK and P-ULK1 in the model group were significantly decreased(P<0.01).Compared with the model group,the protein expression levels of P-AMPK and P-ULK1 in the Shuangshen Huoxue Granules(high,medium and low doses)group and trimetazidine hydrochloride group were increased(P<0.05).The protein expressions of P-AMPK and P-ULK1 in Shuangshen Huoxue Granules(high,medium and low doses)groups were dose-dependent,with statistical significance(P<0.05).Conclusion:1.Yiqi Yangyin Huoxue Tongluo Decoction can improve cardiac function in rats with acute myocardial infarction.2.Yiqi Yangyin Huoxue Tongluo Recipe can up-regulate myocardial AMPK and ULK1 protein expressions in rats with acute myocardial infarction.3.Yiqi Yangyin Huoxue Tongluo Decoction can improve cardiac function in rats with acute myocardial infarction,and its mechanism may be related to AMPK-ULK1 signaling pathway. |
