Objective: To establish an animal model of New Zealand white rabbit for visual fatigue,to evaluate the pharmacodynamic results of the self-prepared "Yiming Tang" to relieve visual fatigue and to explore its mechanism of action,and to provide a theoretical basis and reference value for preclinical pharmacodynamic studies for the development of in-hospital preparations or new drugs for the treatment of patients with visual fatigue.Materials and methods: Twenty healthy New Zealand white rabbits were randomly divided into four groups: blank control group,visual fatigue model group,self-prepared Yiming Tang low-dose group,self-prepared Yiming Tang high-dose group,five rabbits in each group were given the Chinese herbal compound "Yiming Tang" by gavage for 28 days.Yiming Tang" for 28 days.The therapeutic effect of Yiming Tang on New Zealand White rabbits with visual fatigue was measured and analyzed by examining the refractive index,intraocular pressure,anterior chamber depth,and lens thickness of New Zealand White rabbits before and after treatment,and after 28 days,the eyes of the experimental animals were taken,fixed in paraffin,embedded,sectioned,stained with HE,Masson,and α-SMA immunohistochemical staining,and observed by fluorescence microscopy.The pathological changes of the ciliary muscle tissue were observed by fluorescence microscopy.Graph prism 8 was used to statistically analyze the results of the four groups before and after treatment and between the four groups.Results:After 14 days of visual fatigue modeling intervention,the intraocular pressure(P < 0.01)and anterior chamber depth(P < 0.05)were reduced in the model group compared with the normal blank control group,but the refractive index and lens thickness were not statistically different(P > 0.05);after 28 days of visual fatigue modeling intervention with gavage administration,the refractive index and lens thickness were significantly increased(P < 0.01)and reduced(P < 0.05)compared with the normal blank control group.After 28 days of visual fatigue modeling and gavage administration,the refractive index and lens thickness values of New Zealand White rabbits in the model group were significantly higher(P < 0.01)and reduced(P< 0.05)compared with those of the normal blank control group;compared with the visual fatigue model group,the refractive index and lens thickness values of New Zealand White rabbits in the Yiming Tang low-dose administration group were significantly lower(P < 0.01),whereas the refractive index and lens thickness values of New Zealand White rabbits in the Yiming Tang high-dose administration group were lower(P < 0.05).The refractive index of New Zealand white rabbits in the highdose Yiming Tang group was reduced(P < 0.05).Conclusion:1.In this study,a model of New Zealand White rabbits with visual fatigue was established to evaluate the efficacy of self-prepared Yiming Tang,and refractive error values and lens thickness values of New Zealand White rabbits with visual fatigue were improved.2.The self-prepared Yiming Tang could relieve visual fatigue,and after 28 days of treatment,the refractive value and lens thickness of Yiming Tang low-dose group showed significant improvement compared with those before the intervention,and the difference was statistically significant(P < 0.01);the refractive value of New Zealand White rabbits in the low-dose group improved(P < 0.05).
