| Purpose:Focusing on the nature of spleen deficiency in colon cancer patients,we combed through the literature and summarized our clinical practice experience to find the theoretical basis for the treatment of colon cancer with strengthening Qi and splee n to fight cancer Formula and to observe its clinical efficacy.By means of animal ex periments,the anti-inflammatory effect and mechanism of strengthening Qi and spleen to fight cancer Formula on CT26 WT tumour-bearing nude mice were elucidated to pro vide theoretical support for the clinical treatment of colon cancer by strengthening Qi and spleen to fight cancer Formula method.Material and method:Ⅰ.Literature and clinical research:Based on literature research and relevant experimental literature,we investigate the main pathogenesis of colon cancer progression and the application and advantages of traditional Chinese medicine combined with chemotherapy in the treatment of this disease;analyze and summarize the experience of the Department of Oncology of the Affiliated Hospital of Liaoning University of Chinese Medicine(the department)in the treatment of colon cancer,the rules of medication,and form a formula to support the righteousness and drive away the evil for experimental research.Ⅱ.Experimental studies :In this study,SPF-grade BALB/C thymus-free nude mice were used as the initial study subjects.Firstly,20 female and 20 male nude mice of 6 weeks old with body mass(20±5g)were injected with 0.2ml CT26 WT cell suspension subcutaneously in the axilla to prepare a tumor-bearing nude mouse model of colon cancer,and the successfully replicated tumor-bearing nude mice(40 mice)were randomly(random number method)divided into 4 groups,namely: model group,invigorating spleen and replenishing qi decoction combined Chinese medicine treatment group,Capecitabine chemotherapy group,and the group treated with invigorating spleen and replenishing qi decoction combined Chinese medicine combined with capecitabine.The corresponding drug intervention experiments were conducted for the four groups respectively,the material was taken after 15 consecutive days of intervention and observe the general state of the nude mice.The whole blood of colon cancer tumor-bearing nude mice was taken for blood routine examination;the intact tumor tissues were peeled off and weighed,a portion of the tumour tissue was taken and immunoblotting was used relevant proteins(TNF,NF-κBp65,p-NF-κBp65)were detected in the tumor tissues.SPSS 26.0 was used for statistical analysis of the data,and the significant difference was set at P<0.05.Results:Ⅰ.Literature and clinical studies1.The spleen plays an important role in maintaining the normal physiological functioning of the body.The sufficiency of vital energy is due to the absence of injury to the spleen and stomach,which can then nourish the vital energy.If the spleen is deficient,it has no power to transport and transform,so water,grain and essence cannot be transported and transformed,resulting in a lack of nutrients to maintain the normal physiological functions of the body,and thus the vital energy cannot be recharged,which is the cause of all diseases.The deficiency of spleen is also the root cause of tumours.Ultimately,the deficiency of positive qi cannot resist the evil qi,resulting in the invasion of cancer and toxins,and the internal growth of phlegm,dampness,stasis and toxins.2.For many years,the department has been treating colon cancer with formulae based on the principle of helping the righteous and dispelling the evil.The formula for helping the righteous is based on Si Jun Zi Tang,which strengthens the spleen and enhances qi,and is combined with Curcuma longa,Sheng Oyster,Jiang Huang,Xian He Cao and Ban Zhi Lian to dispel the evil,and with Chai Hu,Scutellaria baicalensis and Ban Xia to form a combined formula of Xiao Chai Hu Tang and Si Jun Zi Tang to smoothly connect the combined application of drugs for helping the righteous and dispelling the evil.This formula has been applied to the present day.In this study,the experimental part of the formula was used as a Chinese herbal treatment to benefit Qi and strengthen the spleen against cancer.Ⅱ.Experimental studies1.Body mass of nude rats with tumor: the body mass of nude rats with colon cancer in the capecitabine(chemotherapy)intervention group decreased significantly compared with that before the experiment started,and the difference was statistically significant(P<0.05),while the body mass of nude rats with colon cancer in other groups did not change significantly compared with that before the experiment started,and the difference was not statistically significant(P>0.05).2.Effect of routine blood indicators:Compared with the model group,the hemoglobin content and the number of red blood cells in the peripheral blood of the tumor-bearing nude mice were not statistically different between the groups(P>0.05);Compared with the model group,the number of leukocytes in the peripheral blood of mice in each group was significantly reduced(P<0.01);compared with the strengthening Qi and spleen to fight cancer Formula group,the number of leukocytes in the peripheral blood of mice in the Capecitabine group and the strengthening Qi and spleen to fight cancer Formula combined with Capecitabine group was significantly reduced(P<0.01),and the difference was statistically significant;compared with the Capecitabine group,the number of leukocytes in the peripheral blood of mice in the strengthening Qi and spleen to fight cancer Formula combined with Capecitabine group was significantly increased(P<0.01),and the difference was statistically significant.The difference was statistically significant.3.Tumor weight and tumor suppression rate of nude rats: compared with the control group,the tumor weight of nude rats in each group decreased(P<0.05);the tumor suppression rate was 36.5% in the intervention group of recipe for strengthening Qi and spleen to fight cancer Formula,40.6% in the intervention group of Capecitabine,and 47.4% in the intervention group of recipe for strengthening Qi and spleen to fight cancer Formula combined with Capecitabine.4.TNF-α,p-NF-κBp65,NF-κB p65 protein expression levels:TNF-α,p-NF-κBp65,NF-κB p65 protein expression levels: compared with the model group,TNF-α,p-NF-κBp65,NF-κB p65 protein expression levels were significantly lower in each administration group;compared with the strengthening Qi and spleen to fight cancer Formula group,TNF-α,p-NF-κBp65,NF-κB p65 protein expression levels were significantly lower in the strengthening Qi and spleen to fight cancer Chinese herbal compound combined with the chemotherapeutic drug capecitabine group(P<0.01);compared with the Capecitabine group,TNF-α,p-NF-κBp65,NF-κB p65 protein expression levels were significantly lower in the strengthening Qi and spleen to fight cancer Chinese herbal compound combined with the chemotherapeutic drug Capecitabine group(P<0.01).NF-κB p65 protein expression levels were significantly reduced(P<0.01);compared with the Capecitabine group,TNF-α,p-NF-κBp65,NF-κB p65 protein expression levels were significantly reduced in the strengthening Qi and spleen to fight cancer Formula combined with Capecitabine group(P<0.01),which was statistically significant.Conclusion:1.Through the compilation of TCM literature and clinical practice,it is clear that deficiency of the spleen and resulting weakness of the righteousness and cancer toxicity are important pathogenic mechanisms of malignant colon cancer,and the department can benefit patients clinically by benefiting the qi and strengthening the spleen to fight cancer.2.Benefiting Qi and strengthening the spleen against cancer can interfere with tumour growth to a certain extent and alleviate the side effects of capecitabine treatment in colon cancer tumour-bearing nude mice,and reduce the leucopenia caused by chemotherapy in experimental mice.3.The tumour suppression rate of strengthening Qi and spleen to fight cancer Formula combined with Capecitabine intervention group was better than that of strengthening Qi and spleen to fight cancer Formula group or Capecitabine group.The results of immunoblotting showed that the combination of strengthening Qi and spleen to fight cancer Formula and Capecitabine can down-regulate the expression levels of TNF-α,p-NF-κBp65 and NF-κB p65-related proteins in the tumor tissues of colon cancer tumor-bearing nude mice.The mechanism of action may be through inhibiting the over-activation of NF-κB signaling pathway,down-regulating the expression levels of TNF-α,p-NF-κBp65 and NF-κB p65 proteins and suppressing the inflammatory response in the tumor,so as to achieve the effect of inhibiting tumor development. |
