| Objective:The effective components,target and signal pathway of Wenshen Jianpi recipe(WSJPI)in treating polycystic ovary syndrome(PCOS)were studied from molecular level by using network pharmacological method,and the safety and efficacy of WSJPI recipe were guided.Through clinical research,we observed the clinical effect of Wenshen Jianpi decoction on PCOS of kidneydeficiency and phlegm-dampness syndrome,in order to provide guidance for clinical treatment of PCOS.Part Ⅰ Network Pharmacological Study of Wenshen Jianpi recipe on PCOSMethods:To explore the mechanism of Wenshen Jianpi recipe(WSJPI)in treating Polycystic Ovary Syndrome(PCOS)by network pharmacological method.Methods: Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)was used to Systems Pharmacology the active ingredients and targets of Wenshen Jianpi recipe The targets of PCOS were screened out by human gene database(Genecards)and OMIM database,and the“Drugactive ingredient-target” network was constructed by using Cytoscape 3.9.1 software to screen the core targets Target protein interaction networks were constructed by STRING platform,and Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(Kegg)enrichment analysis were performed using Metascape database.Results:Network pharmacological study obtained 113 active components of Wenshen Jianpi recipe,including Kaempferol,quercetin,isorhamnetin,luteolin,epigallocatechin,etc.,452 drugcomponent targets were obtained,including AKT1,MAPK3,MAPK1,STAT3,PIK3R1 and so on,the specific pathways involved include endocrine resistance,the role of AGE-RAGE signaling in diabetic complications,and Thyroid hormone signaling pathways.Conclusion:The results of network pharmacology showed that Wenshen Jianpi recipe could treat PCOS through multi-component,multi-target and multi-pathway,it plays a therapeutic role by anti-inflammation,regulating glucose and lipid metabolism,regulating endocrine and other ways.Part Ⅱ Clinical Observation on the treatment of PCOS with Wenshen Jianpi decoctionMethods:The patients with polycystic ovary syndrome were selected from the gynecological clinic of Dianjiang County Hospital of traditional Chinese 2021 from September to September 2022,sixty-six patients were randomly divided into two groups: the control group(Daeng 35)and the treatment group(Wenshen Jianpi Fang Group),before and after treatment,the curative effect,TCM syndrome integral,serum hormone level(FSH,LH,T,LH/FSH),endometrial thickness,ovulation rate were compared between the two groups,and the safety indexes were observed.Results:(1)the total effective rate was 93.94% in the treatment group and 72.73% in the control group,with a significant difference between the two groups(P<0.05);(2)the scores of TCM syndrome after treatment in both groups were lower than those before treatment,and the scores of TCM syndrome after treatment in the treatment group were lower than those in the control group,the differences between the two groups were statistically significant(P<0.01);(4)the levels of T,LH,LH/FSH in the two groups after treatment were lower than those before treatment(P<0.01),and the levels of T,LH,LH/FSH in the treatment group after treatment were lower than those in the control group(P < 0.01);(5)there was no significant difference in FSH and E 2 levels between the two groups after treatment(P<0.05);(6)the number of 2-9mm follicles in the two groups after treatment was smaller than that before treatment(P<0.05),and the number of 2-9mm follicles in the treatment group was smaller than that in the control group after treatment.There was a significant difference in the number of 2-9mm follicles between the two groups(P<0.01);Conclusion: The clinical study shows that WENSHENJIANPI prescription can effectively reduce the levels of serum T,LH,LH/FSH,promote the development of follicles,reduce the number of 2-9mm follicles and reduce the TCM syndrome score of patients,improve the quality of life of patients,worthy of further clinical research,promotion and application. |
