Objective:Based on the regulatory effect of SIRT1/PGC-1α signaling pathway on oxidative stress after cerebral hemorrhage,the mechanism of neuroprotective effect of traditional Chinese medicine compound An Nao Ping Chong Recipe was discussed,which provided theoretical support for the clinical treatment of cerebral hemorrhage.Methods:The 20 SD rats were randomly divided into 4 groups,namely:sham surgery group(J group),model group(M group),An Nao Ping Chong Recipe group(A group),and edaravone group(Y group),with 5 rats in each group.The establishment of a rat model of cerebral hemorrhage was achieved by injecting 80 ul of autologous blood into the left caudate nucleus,in which the sham surgical group was not injected wth blood or other reagents.After 3 days of modeling,mNSS scores were performed on each group of rats to assess neurological deficits.Enzyyme-linked immunosorbent assay(ELISA)measured the content of oxidative stress product 8-OHdG in serum to assess the degree of oxidative stress damage in rats.CAT activity kit to detect CAT activity in serum to assess rat antioxidant capacity;Tunel staining was used to observe the apoptosis of neuronal damage in brain tissue after cerebral hemorrhage in rats.SIRT1 and PGC-1α protein expression was detected by immunofluorescence.Results:1.There was no abnormality in the neurological function of rats in the sham surgery group,and compared with the sharn surgery group,the rats in the model group and the drug administration group had obvious neurological function defects(P<0.01);Compared with the model group,the neurological function scores of the An Nao Ping Chong Recipe Group and the Edarafone Group improved significantly(P<0.01).2.Compared with the sham group,the serum 8-OHdG content of rats in the model group was significantly increased(P<0.01),and the CAT activity was also significantly increased(P<0.01).Compared with the model group,the serum 8-OHdG content of rats in the An Nao Ping Chong Recipe and edaravone groups was significantly reduced(P<0.05 or P<0.01),and the CAT activity in the An Nao Ping Chong Recipe group significantly increased(P<0.01).3.TUNEL staining performed on rats with cerebral hernorrhage for 3 days and found that no apoptotic cells were found in the sham surgery group,and obvious apoptosis of nerve cells was seen in the brain tissue around the hemorrhage in the model group and the rats in the administration group(P<0.01);Compared with the model group,the number of neuronal apoptotic cells in the surrounding tissue of the hematoma in the An Nao Ping Chong Recipe group and the edaravone group was significantly reduced(P<0.01).4.Compared with sham surgery,the expression of SIRT1 and PGC-1α in rats in the model group and the drug group was significantly increased(P<0.01).Compared with the model group,the expression of SIRT1 and PGC-1α in the An Nao Ping Chong Recipe Group and the Edarafone Group also increased significantly(P<0.01).Conclusion:An Nao Ping Chong Recipe may play a neuroprotective role by increasing the expression of SIRT1 protein and its receptor PGC-1α protein in the brain tissue surrounding the hemoma of rats with cerebral hemorrhage,reducing the concentration of oxidative stress product 8-OHdG in serum and increasing the activity of antioxidant enzyme CAT,inhibiting neuronal oxidative stress response and apoptosis.
