| Objective: In this study,the possible effect targets of Bielong Ruangan Decoction were screened by network pharmacology,and verified by in vitro experiments to preliminarily explore the mechanism of Bielong Ruangan Decoction in the treatment of primary liver cancer.Methods: Through the database of traditional Chinese medicine and disease,the main effective components and targets of Bielong Ruangan Decoction in the treatment of primary liver cancer were screened by network pharmacology tools,the molecular mechanism of action targets was analyzed,and the relationship network of’ traditional Chinese medicine-component-target-disease’ was constructed.In vitro experiments were used to verify the regulatory effect of Bielong Ruangan Decoction on Hep G2 cells and core targets.Results: A total of 205 potential targets of Bielong Ruangan Decoction in the treatment of liver cancer were screened.The results of enrichment analysis showed that the targets involved a total of 2343 biological processes,108 cell components,267 molecular functions,and 174 signaling pathways.By using the constructed’ Chinese medicine-component-target-disease’ network,it was found that Bielong Ruangan Decoction mainly participated in the treatment of liver cancer by quercetin,glycine and kaempferol on JUN,P53 and ESR1 targets involved in lipopolysaccharide induction,oxidative stress,cell cycle,DNA transcription and other tumor biological behaviors related to liver cancer.In vitro experiments showed that after the intervention of Bielong Ruangan Decoction drug plasma,the cell viability of LX-2/ Hep G2 cells decreased significantly,and the m RNA and protein expression levels of p53 and E2F1 in LX-2/ Hep G2 cells decreased significantly.The expression of LX-2/ Hep G2 cells increased in G1 phase and decreased in S phase.Conclusion: Bielong Ruangan Decoction significantly inhibited the proliferation of Hep G2 cells in vitro,and the specific mechanism was related to the regulation of E2F1 and P53. |
