Effect Of Simo Decoction On The Expression Of CNP And NPR-B In FD Rats With Liver-Spleen Stagnation Syndrom

Objective: To investigate the effects and possible mechanisms of Si Mo Tang in treating functional dyspepsia with liver spleen stagnation,and to provide new ideas and theoretical basis for the prevention and treatment of functional dyspepsia.Methods: Thirty-six SPF grade Wistar rats were randomly divided into a blank group,a model group,a mosapride group and a Si Mo Tang low,medium and high dose group,with six rats in each group.Except for the blank group,the rats in the rest of the groups were prepared with various stimulating factors to create a model of functional dyspepsia with liver and spleen stagnation.After successful modelling,the low,medium and high dose groups were given1.8g/(kg-d),2.4g/(kg-d)and 3.2g/(kg-d)of Si Mo Tang by gavage respectively,while the mosapride group was given 0.305mg/(kg-d)by gavage,and the blank group and the model group were given the same dose of saline once daily for 14 d.After gavage,the rats in each group were weighed and the gastric emptying rate was measured by phenol red method,the small intestine propulsion rate was measured by toner method,the serum MTL and gastrin GAS levels were measured by ELISA,the NPR-B protein content in the gastric sinus was measured by Western-blot,and the CNP m RNA and NPR-B m RNA expression in the gastric sinus was measured by RT-PCR.Results: At the end of gavage,the body weight,gastric emptying rate,small intestine propulsion rate and serum levels of MTL and GAS of the rats in the model group were significantly lower than those in the blank group(P<0.05),and the contents of NPR-B protein,CNP m RNA and NPR-B m RNA in the gastric sinus were significantly higher than those in the blank group(P<0.05);the body weight,gastric emptying rate,small intestine propulsion rate,serum levels of MTL and GAS of the rats in the medium and high dose groups of Si Mo Tang and Mosapride were significantly higher than those in the model group(P<0.05).The body weight,gastric emptying rate,small intestine propulsion rate,MTL and GAS levels in serum were significantly higher than those in the model group(P < 0.05),while the content of NPR-B protein,CNP m RNA and NPR-B m RNA in gastric sinus were significantly lower than those in the model group(P<0.05);the body weight,gastric emptying rate,MTL and GAS levels in serum of rats in the high dose group of Si Mo Tang were significantly higher than those in the low and medium dose groups of Si Mo Tang and Mosapride(P < 0.05).The body weight,gastric emptying rate and serum levels of MTL and GAS in the high-dose group of Si Mo Tang were significantly higher than those in the low and medium-dose groups of Si Mo Tang and the Mosapride group(P<0.05),and the levels of NPR-B protein,CNP m RNA and NPR-B m RNA in the gastric sinus were significantly lower than those in the low and medium-dose groups of Si Mo Tang and the Mosapride group(P<0.05).Conclusion: Si Mo Tang can enhance the gastrointestinal motility of rats with functional dyspepsia due to liver and spleen stagnation,and the mechanism may be related to the increase of serum MTL and GAS levels and the decrease of CNP and NPR-B in the gastric sinus.
