| 1.Objective:In this study,the pathogenesis of KOA was studied based on joint synovial NF-κB signaling pathway by making rabbit KOA model,and relevant indexes of NF-κB signaling pathway in synovial fibroblasts were observed for the prognosis of Zhitongjiangu Fang,so as to clarify whether Zhitongjiangu Fang could regulate the KOA joint synovial NF-κB signaling pathway and achieve therapeutic effect on KOA.It can provide theoretical support and scientific basis for the clinical application of Zhitongjiangu prescription in the treatment of KOA,carry forward the advantages and characteristics of the Chinese medical treatment of KOA,and produce good social and economic benefits.2.Methods:After the successful preparation of the rabbit KOA model,60 healthy adult New Zealand white rabbits were randomly divided into five groups with 12 rabbits in each group,including blank control group,model control group,positive control group,analgesic and bone-strengthening low-dose group and high-dose group according to the principle of body weight balance by random number table method.The positive control group was given cecoxib intragastric administration,while the low-dose and high-dose groups were given Tongzhijiangu decoction intragastric administration.Synovium tissues of rabbit knee joint were taken at three time points before intervention,2 weeks after intervention and 4 weeks after intervention,and the synovium was observed by HE staining.The expressions of IL-1 and MMP-3 in synovium were detected by immunohistochemistry.The expressions of IKKβ and NF-κB p65 were detected by Western Blot.3.Results:Before intervention,inflammatory cell infiltration and fibrous tissue hyperplasia of synovium increased in other groups compared with blank group.At 2 and 4 weeks after intervention,inflammatory cell infiltration was reduced and fibrous tissue hyperplasia was relieved in the drug intervention groups compared with the model group.Compared with blank control group,expressions of IL-1,MMP-3,IKKβ and P65 in synovium of other groups were increased before intervention and at the second week of intervention,with statistical significance(P < 0.05).Compared with model control group,expressions of IL-1,MMP-3,IKKβ and P65 in synovium of positive control group,Zhitongjiangu low-dose group and Zhitongjiangu high-dose group decreased at week 2and week 4,with statistical significance(P < 0.05)4.Conclusion:Zhitong Jiangu decoction can reduce the infiltration of synovitis inflammatory cells and reduce the expression of IL-1,MMP-3,IKKβ and P65 in rabbit KOA model,which may be the mechanism of its action in the treatment of knee osteoarthritis. |
