Objective:To explore the mechanism of the Fas/FasL signaling pathway in a rat model of intervertebral disc degenerative change.We examined the expression levels of Fas/FasL proteins and mRNA expression levels,and then analyzed whether Bushen Huoxue soup could delay lumbar disc degeneration through the Fas/FasL signaling pathway.The results of the study were as follows Thus,we can investigate more precisely the mechanism of the effect of Bushen Huoxue soup to improve the pain and other clinical symptoms caused by lumbar degeneration,discover the potential therapeutic effect of Bushen Huoxue soup,and provide a reliable theoretical basis and exploration direction for the treatment of lumbar intervertebral disc degeneration by Chinese medicine.Methods:The 52 experimental rats were numbered according to their body weight and divided into 3 groups as per After all 3 2 rats in the surgical group had been modeled,2 rats were randomly selected to test the success of modeling,and then randomly divided into 3 groups:Bushen Huoxue soup group,Yaobitong group and the model group,10 rats in each group.Blank group,sham-operated group and model group were given 10mL/kg saline gavage daily;Bushen Huoxue soup group:28.98 g/kg Bushen Huoxue soup daily by gavage;Yaobitong group:0.3 4g/kg Yaobitong capsule daily by gavage.All rats were executed by air embolization after 12h of the last gavage after four weeks of treatment,and the L3/4-L5/6 intervertebral discs were dissected out.They were also used for histomorphological and light microscopic observation and IHC analysis.Western Blot was used to detect Fas/FasL protein expression in five groups,and RT-qPCR was used to detect the mRNA content of Fas/FasL in each group of intervertebral disc cells.Results:The cell morphology of the specimens in the blank and sham-operated groups was basically normal,and the nuclei were clearly visible;the nuclei of the model group were obviously fragmented,with interruptions between the fibrous rings and the nuclei,and the structures were not clear,and the staining was quite dark;Bushen Huoxue soup group showed mild rupture of the fibrous rings,a slight decrease in the number of nucleus cells and vacuoles,and mild condensation of the matrix in the nucleus.Compared with the blank group,Fas/FasL IOD values were significantly higher(P<0.01)and relative protein expression levels and mRNA expression levels were significantly lower(P<0.01)in the model group.The differences in Fas/FasL IOD values,Fas/FasL/β-actin protein expression levels and Fas/FasL mRNA expression levels between the sham-operated group and the blank group were not statistically significant(P>0.05).Compared with the model group,Fas/FasL IOD values were significantly lower(P<0.01),Fas/FasL protein expression levels were significantly increased(P<0.05)and Fas/FasL mRNA expression levels were upregulated(P<0.05)in the Bushen Huoxue soup group and the Yaobitong group.Compared with the Yaobitong group,the Fas/FasL IOD values were significantly lower(P<0.01),and the relative expression levels of Fas/FasL proteins and mRNA expression levels were significantly higher(P<0.01)in the Bushen Huoxue soup group.Conclusion:By being able to effectively treat the damage to the nucleus pulposus and the fibrous ring caused by degenerative lesions of the lumbar spine,Bushen Huoxue soup promote the expression of related proteins,which may be related to its regulation the Fas/FasL signaling pathway.
