| Purpose:In this study,db / m and db / db mice studied the expression level of macrophages and inflammatory factors of abdominal macrophages regulated by MGLL,analyzed and explored the mechanism of its effect on the metabolic reprogramming of macrophages and the treatment of type 2 diabetes.Material and method:8 db / db male db / db / m mice,db / db mice were set as db / db control group(dagliflozgroup),db / db model group,db / db low dose group,db / db sugar dose group,db / db high dose group(28g / kg,14 g / kg,7g / kg,1.29 mg /(kg·d)),db / m mice were set as blank group.After 1 week of adaptive feeding with common feed,gavage administration intervention was performed.The pathological morphology of colon tissue was observed by HE staining;blood from abdominal aorta to explore serum lipid metabolism.The m RNA expression levels of MGLL,2-AG,and CB2 in macrophages of mouse peritoneal adipose tissue were determined by PCR.Protein expression of TLR4,My D88,TRAF6,IRAK4,and phosphorylation of NF-κB p65 \MAPKs P38 in TLR 4 signaling were measured by Western-blot assay.The levels of TNF-α\ TNF-β\ IL-1β\ IL-4 \ IL-6 \ IL-10 in the cell culture supernatants of each experimental group were measured using the ELISA kit.Results:1.It can improve the process of blood glucose and blood lipid metabolism in db / db mice.2.Chinese medicine compound sugar kang can change the histopathological morphology of colon and reduce inflammatory damage in db / db mice.3.Traditional Chinese medicine compound sugar kang can regulate the protein expression of TLR4 signaling pathway through MGLL to cause the process of metabolic reprogramming of macrophages and improve the metabolic syndrome.Conclusion:1.Traditional Chinese medicine compound benefit sugar kang can reduce macrophage protein expression and infiltration of inflammatory factors in abdominal adipose tissue of mice.2.MGLL regulates TLR4 pathway through CB2 to polarization macrophages from M1 to M2 to undergo metabolic reprogramming,reduce insulin resistance,and prevent the occurrence and progression of diabetic disease.3.Traditional Chinese medicine compound sugar kang can inhibit the pathological increase of lipid content in non-adipose tissues in human body,reverse the occurrence of lipid metabolism reprogramming process,prevent the abnormal metabolism of adipose tissue,reduce intestinal inflammatory damage,prevent the occurrence of metabolic syndrome,and thus play a role in preventing diseases. |
