| Purpose:Based on the network pharmacology and molecular docking technology,key components,targets and mechanism of Yixin-Fumai granules in treating sick sinus syndrome and improving anxiety disorder were explored,so as to provide reference for subsequent in-depth research.Material and method:1.The information of active components and related potential targets of Yixin-Fumai granules were obtained through the TCMSP and BATMAN-TCM databases;2.The disease databases of Gencards,OMIM and Dis Ge NET were used for screen the relevant targets of sick sinus syndrome and anxiety disorder,and the common targets of benefiting cardioverter granule,sick sinus syndrome and anxiety disorder were collected;3.The network of active components-target-disease was constructed by Cytoscape software,and the key components and core targets of Yixin-Fumai granules for the treatment of sick sinus syndrome with anxiety disorder were obtained by topology analysis with Cyto NCA plug-in;4.The molecular mechanism of Yixin-Fumai granules in the treatment of sick sinus syndrome complicated with anxiety disorder was obtained through the enrichment analysis of common targets through DAVID database;5.Molecular docking is conducted through Auto Dock Tools software to verify whether the combination of key components and core targets is reliable.Results:1.173 effective components of Yixin-Fumai granules in the treatment of sick sinus syndrome complicated with anxiety disorder were predicted,and 10 key chemical components such as quercetin,deoxyharpertonine and luteolin were obtained by topological analysis.STAT3,VEGFA,PTGS2 and other 10 core targets.2.Through enrichment analysis,we speculated that the mechanism of benefiting heart and pulsing granule heteropathy in the same treatment of SSS with ad might be:(1)Regulating cytokine activity and delaying myocardial fibrosis;(2)Reduce ischemia,inflammation,apoptosis and other damage to sinoatrial node cells and neurons;(3)Delay the induction and aggravation of SSS and AD caused by diabetes,arteriosclerosis,tumor and virus infection.3.Molecular docking confirmed that nine core targets except CASP3 had strong binding activity with key components.Conclusion:In this study,network pharmacology and molecular docking were used to analyze and validate Yixin-Fumai granules for the treatment of anxiety disorders associated with sick sinus syndrome,and we initially predicted the dominant components and mechanism of action of Yixin-Fumai granules for the treatment of anxiety disorders associated with sick sinus syndrome,and provided a theoretical support for the treatment of sick sinus syndrome associated with anxiety disorders associated with sick sinus syndrome with complex cardiac abnormalities. |
