Objective: To observe the effect of renewing and joining solution on tendon bone healing after rotator cuff injury repair in rabbits.Methods: A random number table was used to divide 36 adult healthy New Zealand rabbits into three groups,12 rabbits in each group as the sham-operated group,the experimental group and the model group,respectively.In the sham-operated group,only skin incision and suturing were performed,and no rotator cuff injury was modeled,and gavage was administered with equal doses of saline;in the experimental group,rotator cuff reconstruction surgery was performed at 1 week after modeling,and gavage was administered with a bone repair solution;in the model group,rotator cuff reconstruction surgery was performed at 1 week after modeling,and gavage was administered with saline.The duration of treatment was 4 weeks,and the patients were executed and removed at the end of the treatment period.Histological staining,Western Blot for transforming growth factor-β1 and platelet-derived growth factor-BB and ELISA for relevant inflammatory indexes were observed.Results:(1)Histopathology: in the sham-operated group(K),the tendon-bone surface was tightly connected,the interface was clear,the tendon fibers were neatly arranged in an axial shape,the tendon cells between the fiber bundles were regularly arranged among them,the chondrocytes were in the same direction as the muscle fibers,the whole structure was clearly visible,and there were no pathological changes.In the experimental group(S),the tendon-bone surface was tightly connected,and the interface was blurred,but still visible.There were a few inflammatory cells in the tendon-bone junction,and there was visible neovascularization,as well as a large number of chondrocytes and a small amount of calcified cartilage production.In the model group(M),the tendon-bone surface was loosely united,with a small amount of union between the tendon breaks and a large number of inflammatory cells visible,without obvious capillaries,and with increased fibroblasts and irregular arrangement.(2)ELISA assay: T he concentrations of IL-6 and TNF-α in the model group and the tendon-reinforcing fluid group were higher than those in the sham-operated group,P<0.05,which was statistically significant,and the concentrations of IL-6 and TNF-α in the tendon-reinforcing fluid group were lower than those in the model group,P < 0.05,which was statistically significant.(3)The relative expression levels of TGF-β1 and PDGF-BB proteins in the rotator cuff cartilage tissues of the three groups after 4 weeks of administration were detected by Western Blot.Compared with the sham-operated group,the expression levels of TGF-β1 and PDGF-BB proteins were upregulated in the model group and the renewed tendon and bone solution group(P < 0.01;P<0.05);compared with the model group,the expression levels of TGF-β1 and PDGF-BB proteins were up-regulated in the sequestra bone-setting solution group(P<0.01),and the differences were statistically significant,indicating that the sequestra bone-setting solution could significantly promote the expression of TGF-β1 and PDGF-BB protein expression.Conclusion: The possible mechanism by which the tendon and bone healing after rotator cuff injury repair can be promoted by the renewal solution is related to its reduction of inflammatory factors such as TNF-α and IL-6 and regulation of TGF-β1 and PDGF-BB protein expression.
