| Objective:In this study,the effects of acupuncture and Buy ang Huanwu Tang(BYHWT)on cerebral infarction area,brain tissue water content,blood-brain barrier permeability and tight junction proteins occloudin and claudin-5 in MCAO model rats were observed,and part of the therapeutic mechanism of acupuncture and drug combined with acupuncture on cerebral infarction was explored.Methods:SPF-grade SD rats were randomly divided into sham surgery group and surgical module,and the model of cerebral infarction caused by middle cerebral artery embolization was selected,and the model rats were randomly divided into model group,acupuncture group,drug group and acupuncture group for the second time.The acupuncture group selected the acupoints of "DU26,PC6,SP6",the drug group was given to BYHWT for gastric gavage,the acupuncture group and the sham surgery group were acupuncture and supplement yang to return five soups for gastric gavage,the model group only grabbed and fixed without intervention,acupuncture and fixation operations were performed for 20min each time,twice a day,and Chinese medicine gavage 0.6ml/100g/only administration,once a day,for 5 days.After the intervention,TTC staining was used to detect the infarct area of brain tissue,dry and wet weight to detect the water content of brain tissue,Evans blue(EB)method to detect BBB permeability,and Western blot to detect occludin and claudin-5 expression levels.Results:1.The area ratio of cerebral infarction:There was no significant difference among the model group,acupuncture group,drug group,acupuncture and drug group at 3 days after surgery(P>0.05).5 days after surgery,the area of cerebral infarction in the acupuncture and drug group was significantly reduced compared with the acupuncture group(P<0.05).2.The content ratio of Water in brain tissue:Compared with the sham surgery group,the water content of brain tissue in the model group increased(P<0.01)at 3d and 5d after surgery,and there was no difference in water content among the model group and acupuncture group,and the drug group in 3d after surgery(P>0.05).The water content of brain tissue in the acupuncture and drug group was significantly reduced compared with the model group(P<0.0 5),Compared with the acupuncture group and drug group,the water content of brain tissue was reduced in the acupuncture group and drug group,and the difference was not obvious(P>0.05).At 5 days after surgery,there were no significant differences among the model group,acupuncture group,drug group,acupuncture and drug group(P>0.05).3.The EB detection in brain tissue:Compared with the sham surgery group,the EB content was increase in the model group at 3d and 5d after surgery(P<0.01).Compared with the model group,there were no significant differences among the model group,acupuncture group,drug group,acupuncture and drug group(P>0.05).the EB content in the acupuncture and drug group was less than acupuncture group and drug group,the differences were no significant at 3 days after surgery(P>0.05).Compared with the model group,the EB content was decreased significantly in the acupuncture group,drug group,acupuncture and drug group at 5 days after surgery(P<0.01),and there was no significant change among groups(P>0.05).4.The expression detection of occludin,claudin-5 in Blood-brain barrier:(1)The expression detection of occludin:3 days after surgery,compared with the sham surgery group,there was no significant difference of occludin content in the model group(P>0.05);Compared with the model group,there was no significant difference in the acupuncture and drug group(P>0.05).5 days after surgery,compared with the sham surgery group,the expression of occludin in the model group increased(P<0.05),the expression level of occludin protein in the acupuncture group was increased significantly compared with the model group(P<0.0 1),and there was no significant difference between the acupuncture group,drug and acupuncture group(P>0.05).②The expression detection of claudin-5:Compared with the sham surgery group,there was no significant difference in the content of claudin-5 in the brain tissue of rats in the model group(P>0.05);3 days after surgery,there was no significant difference between the model group,acupuncture group,drug and acupuncture group(P>0.05);5 days after surgery,compared with the acupuncture group and drug group,the content of claudin-5 in the acupuncture and drug group has upward trend,there was no significant difference among groups(P>0.05).Conclusion:1.Xingnao kaiqiao acupuncture and BYHWT can effectively reduce the area of cerebral infarction in rats.2.Xingnao kaiqiao acupuncture and BYHWT can reduce the cerebral edema of rats.3.Xingnao kaiqiao acupuncture and BYHWT can reduce the permeability of the blood-brain barrier.4.Xingnao kaiqiao acupuncture and BYHWT may reduce BBB damage by promoting the expression of blood-brain barrier tight junction proteins occludin. |
