Objective: To explore the mechanism of tiebaojin cataplasm in the treatment of lumbar disc herniation based on the signal pathway of nuclear factor κ B(NF-κB).Methods: Forty-six SPF 3-month-old male rats were randomly divided into normal group(N =10).The other were used to establish Lumbar disc herniation(LDH)rat model by Autologous nucleus pulposus transplantation,After successful modeling,the rats were randomly divided into model group,tiebaojin cataplasm group,positive drug group,10 rats in each group,The normal group and the model group were smeared L 5 paravertebral with normal saline,once a day for 30 days.tiebaojin cataplasm group was treated with 1.54 cm × 2.09 cm plaster,once a day for 30 days.The positive drug group was treated with voltaren ointment,once a day for 30 days.After30 days,fasting for 12 hours,anesthetized rats,abdominal aorta blood collection,The right l 5nerve root was exposed,and the compressed nerve root was harvested from the cryopreserved platform and the rats were killed.The nerve function score of rats were determined;The histomorphological changes of dorsal root ganglion(DRG)were detected by hematoxylin eosin(HE)staining;The levels of interleukin-1β(IL-1β),interleukin-1β(IL-6),tumor necrosis factor-α(TNF-α)were detected by enzyme-linked immunosorbent assay(ELISA).The m RNA expression levels of Toll-like receptor 4(TLR4),Myeloid differentiation factor(My D88),NF-κ Bp65 were detected by real-time quantitative polymerase chain reaction(real-time PCR).Results:(1)The results of nerve function score showed that Compared with the model group,the scores of tiebaojin cataplasm group and the positive drug group decreased at the 10 th,20th and 30 th day after intervention(P<0.05).(2)According to HE staining,compared with model group,the edge and nucleolus of nucleus were clearer in tiebaojin cataplasm group and positive drug group.Nissl bodies in cytoplasm were arranged more evenly,and vacuoles in cytoplasm were reduced in tiebaojin cataplasm group and positive drug group,and the positive drug group showed more significant improvement.(3)As revealed by ELISA results,compared with normal group,the model group showed increased serum levels of IL-1β,IL-6,TNF-α(P<0.05),while compared with the model group,all the groups with drug intervention showed decreased levels of IL-1β,IL-6,TNF-α(P<0.05).(4)Realtime PCR results showed that compared with the normal group,the model group showed increased relative m RNA expression of TLR4,My D88,NF-κ Bp65(P< 0.05),while compared with the model group,all the groups with drug intervention showed decreased relative m RNA expression of TLR4,My D88,NF-κ Bp65(P<0.05).(5)Immunohistochemistry showed that,The expression levels of TLR4,Myd88 and NF-κBP65 in the model group,tiebaojin cataplasm group and positive drug group were significantly higher than those in the normal group(P < 0.05),the expression levels of TLR4,Myd88 and NF-κBp65 positive cytokines in tiebaojin cataplasm group and positive drug group were decreased(P < 0.05).Conclusion: Tiebaojin cataplasm can reduce the release of inflammatory factors,decrease the score of nerve function,affect the metabolism of NF-κB signal pathway,down-regulation the expression of P65,TLR4,Myd88 m RNA,and relieve the symptoms of lumbar disc herniation.Therefore,tiebaojin cataplasm may be useful in inhibiting inflammatory factors and treating lumbar disc herniation.
