| Objective: In this study,we investigated the improvement effect of Xinjian granules in regulating low mood and its possible mechanism of action,in order to provide a theoretical basis for the development of a functional food that is safe,effective and can regulate low mood.Methods: Under the same environmental conditions,the effect and mechanism of Xinjian granules on regulating mice mood were studied by acute model and chronic stress model respectively.1.Screening of different doses of Xinjian granules using acute depression model(tail suspension test,TST;forced swimming test,FST)and reserpine-induce depression model.2.By establishing chronic sleep interruption model(SI)to induce depression-like behavior,the effect of Xinjian granules on the improvement of mood depression caused by chronic stress model was evaluated by using open field test(OPT),TST,FST and elevated plus maze test(EPMT),followed by related mechanism studies.3.By establishing a chronic social defeated stress model(CSCD),the behavioral assays such as social interaction test(SIT),sucrose preference test(SPT),OPT and TST were used to evaluate the ameliorative effect of Xinjian granules on chronic stress model-induced depressed mood,followed by related mechanism studies.Results: 1.In the acute depression model,the results showed that Xinjian granules 0.3,0.6 and1.2 g/kg significantly reduced the immobility time in FST and TST,which is stable and effective.It also effectively improved the ptosis in mice caused by reserpine injection,proving that Xinjian granules had an ameliorating effect on depression-like behavior in mice and the reserpine-induced depression model.2.In the chronic sleep interruption model,each dose of Xinjian granules(0.3,0.6,1.2g/kg)significantly reduced the immobility time in TST and FST(P<0.05),and the 0.3,0.6 g/kg of Xinjian granules significantly increased open/total arms entries ratio and open/total arms time ratio in EPMT(P<0.05).After the administration intervention,it significantly reduced the serum CORT level(P<0.05)and increased the 5-HT,5-HIAA,NE,E,DA and GABA levels in the hippocampus and cortex of mice(P<0.05).3.In the chronic social defeated stress model,each dose of Xinjian granules(0.3,0.6,1.2 g/kg)improved social interaction behavior and significantly reduced the immobility time in TST and FST(P<0.05);0.6,1.2 g/kg of Xinjian granules significantly reversed the decrease in sucrose preference index caused by CSDS stress(P<0.05);1.2 g/kg of Xinjian granules shortened the feeding latency in the novelty-suppressed feeding test(NSFT)(P<0.05).After the drug administration intervention,it significantly reduced the serum CORT level in mice(P<0.05)and elevated the 5-HT,NE,DA,levels in the hippocampus and cortex of mice(P<0.05).Conclusion: Xinjian granules can play a role in regulating mood by regulating the levels of CORT,5-HT,NE and DA.It is speculated that the mechanism may be related to the inhibition or regulation of neurotransmitter content by HPA.As a medicinal product,Xinjian granules are not only safe and effective for long-term use,but also provide an important basis for the development of health care products to regulate low mood. |
